摘要:

利用从老芒麦(Elymus sibiricus)自身基因组中开发出来的6对多态性SSR引物,对3个老芒麦牧草品种和7个种质进行分子指纹图谱鉴别研究.结果表明:6对SSR引物在这10个材料中共检测出21个多态性SSR标记片段,每对引物可以检测到2~5个数目不等的片段,平均为3.5个.6对引物中3对核心引物组合可以将这10个材料进行有效鉴别.进一步选用3对引物对2个老芒麦品种进行纯度鉴定,表明SSR标记可以对品种群体中具有变异位点的个体进行有效鉴定,同时揭示野生栽培品种‘同德’老芒麦比育成品种‘多叶'老芒麦具有较高的遗传变异度.

Abstract:

Molecular fingerprinting maps of 3 Elymus sibiricus cultivars and 7 germplasms were carried out using 6 simple sequences repeat (SSR) markers developed from the genomic DNA of E. sibiricus. A total of 21 polymorphic SSR fragments were detected in all the tested materials, and 2 to 5 alleles were detected by each pair of SSR primer with an average of 3.5. All the 3 cultivars and 7 germplasms could be well distinguished by the fingerprinting maps constructed by 3 pairs of core primers. In addition, seed purity assessment was conducted in cultivars ‘Tongde' and ‘Duoye' with 3 pairs of SSR primers. It showed that seed purity and the polymorphism of SSR alleles could be effectively identified. Moreover, it revealed that ‘Tongde', a domesticated cultivar, had much lower purity and higher polymorphic alleles than ‘Duoye', a breeding cultivar. The SSR markers used in this study can be a powerful tool for cultivars or germplasms identification, seeds purity assessment in E. sibiricus.
