直立型扁蓿豆成熟胚离体培养器官发生组织学研究

摘要: 本文首次报导了直立型扁蓿豆(Melilotoides ruthenicus (L.) Sojakcv.'Zhili')成熟胚离体培养中器官发生的组织学研究。选用MS培养基并附加不同浓度的外源激素MA、NAA和BA进行培养,定期做冰冻切片进行观查。结果表明:IAA、NAA与BA的配合使用对直立型扁蓿豆成熟胚愈伤组织的诱导和不足芽的器官发生有显著的促进作用。不定芽起源于胚轴的皮层细胞,或表皮和皮层细胞同时启动。愈伤组织的形成可分为三个时期,启动期、分裂期、形成期;不定芽形成全过程可分为四个阶段:启动阶段、芽原基形成阶段、叶原基形成阶段和不定芽形态建成阶段。

Abstract: The histology of organogenesis in isolated culture of mature embryo of Melilotoides ruthenicus (L.) Sojak cv.' Zhili' was reported in this paper.MS medium with IAA, NAA and BA in different concentrations for culturing and freezing slices at different times were observed.The results showed that the use of combination of IAA, NAA and BA had sighficant effect on inducing the callus from mautre embryo and promoting the organogenesis of adventitious buds of the plants.Adventitious buds derived from the cortex cells.or both cutex and coriex cells of blastoaxis.The formation course of callus can be di-vided into 3 stages, i.e., initial stage, fission stage and formation stage.The formation course of adventi-tious buds can be divided into 4 stages:i.e., initial stage, bud promordium formating stage, leaf primordi-um formating stage and bud morphogenesis stage.
