刺五加不同产地种子生理差异的多维度解析

Multidimensional Analysis of Physiological Differences in Seeds of Acanthopanax senticosus from Different Origins

摘要: 为阐明不同产地刺五加(Acanthopanax senticosus)种子萌发差异的生理机制,本研究以黑龙江省阿城区(AC)、木兰县(ML)、方正县高楞镇(GL)和通北县(TB)4个产地种子为材料,分析其表型性状、萌发率、沙藏过程中内源激素动态变化及代谢组特征。结果表明,GL产地种子萌发率和百粒重显著高于其他产地。激素水平上,GL种子沙藏末期油菜素内酯(Brassinosteroid,BR)含量最高、脱落酸(Abscisic acid,ABA)含量最低。代谢组学结果显示,GL种子沙藏前富集糖类并下调胁迫相关代谢物,沙藏后富集能量供应与物质合成相关代谢物。加权基因共表达网络分析(Weighted Gene Co-expression Network Analysis,WGCNA)表明,沙藏前MEturquoise和MEred模块与百粒重显著正相关;沙藏后,MEyellow模块与萌发率呈显著正相关,明确了关键调控代谢物。综上,GL产地种子凭借优良表型、激素平衡与高效代谢表现出更强萌发潜力。研究为优化沙藏技术与提高人工繁育效率提供理论依据。

Abstract: To elucidate the physiological basis underlying provenance-dependent variation in seed germination of Acanthopanax senticosus, seeds collected from four locations in Heilongjiang Province, China-Acheng District (AC), Mulan County (ML), Gaoleng Town of Fangzheng County (GL), and Tongbei County (TB)-were examined for phenotypic traits and germination performance, together with endogenous hormone dynamics during sand stratification and metabolomic profiles. The results showed that seeds from GL exhibited significantly higher germination rates and greater hundred-seed weight than those from the other provenances. In terms of hormonal regulation, GL seeds displayed the highest brassinosteroid (BR) content and the lowest abscisic acid (ABA) content at the late stage of stratification. Metabolomic analysis further indicated that prior to stratification, GL seeds were characterized by an accumulation of carbohydrates accompanied by reduced levels of stress-associated metabolites, whereas after stratification they preferentially enriched metabolites related to energy supply and biosynthetic processes. Weighted gene co-expression network analysis (WGCNA) revealed that the MEturquoise and MEred modules were positively associated with hundred-seed weight before stratification, while the MEyellow module was correlated positively with germination rate after stratification, highlighting key metabolite regulators. Collectively, these findings suggested that the superior germination potential of GL seeds was attributable to favorable seed traits, a more balanced hormonal status, and metabolically efficient reprogramming. This study provides a theoretical foundation for optimizing sand stratification practices and improving artificial propagation efficiency.
