疯草内生真菌FEL3生产SW培养基的最佳碳氮比优化

Abstract: Effect of C/N ratio in culture medium on producing Swainsonine (SW) using Oxytropis kansuensis endophytes FEL3 was studied by Central Composite Design (CCD).The best carbon source and nitrogen source were determined through single factor experiment.Results showed that sucrose and peptone were the best carbon and nitrogen source respectively.The predictive polynomial quadratic equations model was obtained by Design-Expert 7.1.6 software.When sucrose and peptone were 1.47 g·L-1and 0.29 g·L-1 respectively,the SW content was maximum, (6.71×10-4 mol·L-1).These results provide technical support for large-scale SW production using Oxytropis kansuensis endophytes FEL3.

摘要: 为探讨碳氮比对甘肃棘豆(Oxytropis kansuensis)内生真菌FEL3产苦马豆素(Swainsonine,SW)的影响,利用单因素试验对最佳碳源和氮源进行了筛选。结果表明:蔗糖和蛋白胨分别是理想的碳源和氮源;采用中心组合设计(Central Composite Design,CCD)考察液体培养基中蔗糖和蛋白胨的比率对内生真菌FEL3产SW的影响,并利用Design-Expert 7.1.6软件模拟得到二次多项式回归方程的预测模型;当蔗糖为1.47 g·L-1,蛋白胨为0.29 g·L-1时,SW浓度达到最大值6.71×10-4mol·L-1。试验结果将为利用疯草内生真菌FEL3批量生产SW提供技术支持。
