基于DNA宏条形码技术分析布氏田鼠的食物组成及其季节差异

Abstract: Brandt’s vole (Lasiopodomys brandtii) is an important rodent species in typical grasslands, and the analysis of its dietary can deepen the understanding of the foraging strategy of Brandt’s vole and provide a certain reference for the rational management of Brandt’s vole population. In this study, we collected 20 (10♀, 10♂) stomach contents per month from adult Brandt’s voles in May, July, September, and November 2022 in typical grasslands of Dongwuzhumqin Banner, Xilingol League, Inner Mongolia, respectively, for a total of 80 (40 ♀, 40 ♂) stomach contents in 2022. Seasonal differences were analyzed using DNA macro-barcoding technology for dietary identification. The results showed that: (1) a total of 16 plant species from 9 families and 14 genera were identified in the stomach contents of Brandt’s voles. (2) there was no significant seasonal difference in the diet diversity of Brandt’s voles, but the diet richness was significantly higher in autumn than in summer (P<0.01) and winter (P<0.05). (3) There were significant seasonal differences in the proportions of Brandt's voles feeding on Astragalus membranaceusAllium polyrhizumCleistogenes squarrosaChloris virgataPotentilla bifurca and Parthenocissus tricuspidate P<0.05). There were highly significant seasonal differences in feeding on Oxybasis glaucaLeymus chinensisEragrostis minor and Allium ramosum P<0.01).

摘要: 布氏田鼠(Lasiopodomys brandtii)是典型草原的主要鼠种,针对其开展食性分析能够加深对布氏田鼠采食策略的理解,为合理管理布氏田鼠种群提供一定的参考。本研究在内蒙古锡林郭勒盟东乌珠穆沁旗典型草原,分别于2022年5、7、9、11月,每月采集布氏田鼠成体胃内容物20份(10♀,10♂),共计80份(40♀,40♂)胃内容物。利用DNA宏条形码技术进行食性鉴别,分析其季节性差异。结果发现:(1)布氏田鼠胃内容物中共鉴定出植物9科14属16种;(2)布氏田鼠食物多样性无显著的季节间差异,但秋季食物丰富度显著高于夏季(P<0.01)和冬季(P<0.05);(3)布氏田鼠对黄芪(Astragalus membranaceus)、多根葱(Allium polyrhizum)、糙隐子草(Cleistogenes squarrosa)、虎尾草(Chloris virgata)、二裂委陵菜(Potentilla bifurca)和地锦(Parthenocissus tricuspidata)的取食比例存在显著的季节差异(P<0.05)。对灰绿藜(Oxybasis glauca)、羊草(Leymus chinensis)、小画眉草(Eragrostis minor)和野韭(Allium ramosum)的取食存在极显著的季节差异(P<0.01)。
