青海野生草地早熟禾响应低温胁迫的代谢组学研究

Metabonomics Study of Qinghai Wild Poa pratensis in Response to Low Temperature Stress

摘要: 为探究青海野生草地早熟禾(Poa pratensis)对低温胁迫的响应机制,本试验以青海省野生草地早熟禾抗寒材料‘10-122’和低温敏感材料‘09-126’为研究对象,采用生理学测定方法及气相色谱-质谱联用技术(GC-MS)分析了低温胁迫下叶片的生理响应及代谢产物差异。试验结果表明,低温敏感材料‘09-126’在低温胁迫下过氧化氢、丙二醛含量和超氧根自由基离子生成速率均显著增加,氧化损伤程度高于‘10-122’;低温胁迫下,‘09-126’和‘10-122’氨基酸含量分别比常温处理提高了1.91倍和1.97倍,可溶性糖含量分别比常温处理提高了4.83倍和2.81倍。采用代谢组学的方法进一步对差异代谢产物进行分析,发现低温胁迫下,‘09-126’中差异代谢物有116个上调,30个下调,‘10-122’有66个上调,68个下调,差异代谢物主要富集在糖及糖醇代谢、氨基酸代谢和三羧酸循环代谢通路中,其中脯氨酸、亮氨酸、鸟氨酸和苏氨酸可以作为草地早熟禾响应低温胁迫的主要标志物,丙氨酸、天冬氨酸和丝氨酸含量的变化可能是‘10-122’抗寒性高于‘09-126’的主要原因。低温条件下‘09-126’中棉子糖含量的增加以及‘10-122’中海藻糖-6-磷酸含量的增加可能是其适应低温的主要机制。

Abstract: In order to clarify the response mechanism of Qinghai wild kentucky bluegrass (Poa pratensis) to low temperature stress,this experiment took cold resistant germplasm material ‘10-122’ and low temperature sensitive germplasm material ‘09-126’ as research objects,the physiological responses and metabolite differences of leaves of two wild P. pratensis under cold stress were analyzed by physiological methods and GC-MS. The results showed that the content of H2O2,MDA and production rate of O2·- in low temperature sensitive ‘09-126’ increased significantly,and the degree of oxidative damage was higher than that in ‘10-122’. Under low temperature stress,the amino acid content of ‘09-126’ and ‘10-122’ was 1.91 times and 1.97 times higher than that of normal temperature,respectively. And the soluble sugar content was 4.83 and 2.81 times higher than that of normal temperature,respectively. Further analysis of differential metabolites by metabonomics showed that under low temperature stress,116 metabolites of ‘09-126’ were up-regulated,and 30 metabolites were down regulated;66 metabolites of ‘10-122’ were up-regulated and 68 metabolites were down regulated. The differential metabolites were mainly concentrated in the metabolism of sugar and sugar alcohol,amino acid and TCA cycle. Among them,proline,leucine,ornithine and threonine can be used as the main markers of Poa pratensis in response to low temperature stress. The change of alanine,aspartic acid and serine content may be the main reason why the cold resistance of ‘10-122’ material is higher than that of ‘09-126’ material. The increase of the content of raffinose in ‘09-126’ and trehalose-6-phosphate in ‘10-122’ may be the main mechanism of their adaptation to low temperature.
