8个小麦条锈菌生理小种产孢相关基因PsCon1克隆及表达分析

Cloning and Expression Analysis Conidiation-Related Gene Pscon1 from Eight Physiological Races of Puccinia striiformis f.sp.tritici

摘要: 为研究基因Pscon1在不同小种中核苷酸序列及相对表达量差异,以小麦条锈菌(Puccinia striiformis f. sp. tritici)基因组DNA为模板,在小麦条锈菌8个生理小种中分别克隆产孢相关基因Pscon1,并对其核苷酸和氨基酸序列进行比对及相对表达量分析。结果表明:在8个小种中分别克隆到条锈菌产孢相关基因Pscon1,基因大小为531 bp,由3个外显子和2个内含子构成,开放阅读框长252 bp,不同小种基因Pscon1核苷酸序列只在44,65,331,465,495和510 bp位点处发生突变,均未引起氨基酸序列变化,且不同生理小种Pscon1在接种'铭贤169' 12 h后相对表达量有差异,水源11-3最高,最低的是Hybrid46-8。推测基因Pscon1在小麦条锈菌不同生理小种产孢过程中相对表达量有差异,这将为进一步研究产孢量差异因素提供参考。

Abstract: In order to research the difference of nucleotide and relative expression level between different races, Puccinia striiformis f. sp. tritici was used as the DNA source. Conidiation-related gene Pscon1 was cloned and isolated from eight epidemic physiological races, and nucleotide and amino acid level and relative expression levels were analyzed. Results indicated that a 531 bp DNA sequence of PsConl was cloned and comprised three extrons and two introns,including a complete 252 bp open reading frame. Sequence analysis indicated that the PsConl sequences of tested physiological races had a few mutations in the site of 44 bp, 65 bp, 331 bp, 465 bp, 495 bp and 510 bp, but no amino acid sequences were changed. The relative expression levels of tested physiological races were different after 12 h inoculation. Accession Su11-3 had the greatest expression while Hybrid46-8 had the least among tested physiological races. It was concluded that the effect of conidiation-related gene Pscon1 on the spore production of different physiological races was different.
