箭叶橐吾内生真菌分离鉴定及组织分布特性研究

Isolation,Identification and Tissue Distribution Characteristics of Fungal Endophyte in Ligularia sagitta

摘要: 为探明有毒植物箭叶橐吾(Ligularia sagitta)的内生真菌种类及其组织分布特性,本试验对植物样品进行表面消毒后分离其内生真菌,采用形态学和内转录间隔区(Internal transcribed spacer,ITS)序列分析技术对分离到的菌株进行种属鉴定并构建系统进化树。结果显示,从箭叶橐吾中分离到21株内生真菌,属于3纲、3目、7科、9属。根据亲缘关系,将其主要分为种群I[以柄孢壳菌属(Podospora sp.)为代表]和种群II[以交链孢霉属(Alternaria sp.)为代表],其中交链孢霉属和柄孢壳菌属均占分离菌株的19.05%,为其优势菌属,主要分布于花和叶。箭叶橐吾的内生真菌在花中分布最普遍(占分离菌株的42.86%),其次为叶(占分离菌株的38.10%)、茎(占分离菌株的14.29%)和根(占分离菌株的4.76%)。结果表明,内生真菌在箭叶橐吾中的分布存在明显组织差异。

Abstract: In order to explore the endophytic fungi species and tissue distribution charateristics of Ligularia sagitta, endophytic fungi were isolated from it using surface disinfection, and morphology and ITS sequence were used to identify the species of endophytic fungi and construct phylogenetic tree. The results showed that 21 strains of endophytic fungi were isolated from Ligularia sagitta, which belonged to 3 classes, 3 orders, 7 families, and 9 genera. They were mainly divided into group I (the representative of Podospora sp.) and group II (the representative of Alternaria sp.) according to their phylogenetic relationship. Among them, Alternaria sp. and Podospora sp. were the dominant species, which were accounted for 19.05% of the isolated strains and mainly distributed in flowers and leaves. Endophytic fungi were most commonly distributed in flowers of Ligularia sagitta (accounting for 42.86% of the isolates), followed by leaves (38.10%), stems (14.29%) and roots (4.76%). The results indicated that the distribution of endophytic fungi existed obvious differences in various tissues of Ligularia sagitta.
