超薄层等电聚焦电泳技术检验燕麦种子的真实性

Abstract: Seed prolamine variation among sixteen oat(Avena spp.)seed samples,consisting of nine varieties and four samples of unknown names cultivated in Qinghai Province,and three varieties cultivated in Hebei Province,was analyzed using ultrathin-layer isoelectric focusing electrophoresis(UTLIEF).The results show that among the tested varieties,a total of 34 bands were identified.Based on distinctive bands at PI point of 4.8,6.55,6.9,7.7,8.4,and 8.6,respectively.Thirteen of the 16 tested samples differentiated from each other.In the present paper,the whole seed,1/2 seed units and 1/4 seed units of Qingyongjiu No.473,Canadian oat,Bayou No.5,and U4,as well as the seeds of Canada oat after aging treatment were electrophoresed by UTLIEF.The results prove that seed variability and damaged seeds of half of their original size,or even dead seeds did not affect the electrophoresis results.

摘要: 利用种子醇溶蛋白超薄层等电聚焦电泳(UTL IEF)技术对青海和河北省燕麦属(Avena spp)16个品种真实性进行了比较分析。结果表明,参试样品的种子醇溶蛋白UTL IEF电泳图谱共分离出34条不同迁移率的谱带;依据蛋白电泳图谱等电点(P I值)为4.8、6.55、6.9、7.7、8.4和8.6等位置谱带的有无以及数目,可鉴别16个参试样品中的13个样品;对同一样品的整粒种子、1/2粒和1/4粒种子,以及人工老化处理获得的同一样品不同发芽率的种子进行超薄层等电聚焦电泳,结果表明破损程度到1/2粒种子和不同生活力种子甚至死种子对电泳鉴定结果基本没有影响。
