多孢木霉HZ-31菌株胁迫下野燕麦内参基因的选择

Abstract: In order to construct a RT-qPCR system suitable for stress of Trichoderma polysporum HZ-31 to wild oats (Avena fatua L.), and to screen stable internal reference genes, GeNorm, NormFinder, BestKeeper and RefFinder were used to analyze 8 candidates of internal reference genes (18S, 28S, TUA, UBC, ACT, GAPDH, TBP and EF-1α) in A. fatua after inoculation of T. polysporum HZ-31. The results showed that TBP, 18S and UBC were the most stable internal reference genes, TBP and TUA, TBP and GAPDH, 18S and TBP, UBC and 18S were the most suitable combination of the two internal reference genes, which could be used as the internal reference genes for functional gene expression analysis during the interaction between T. polysporum HZ-31 and A. fatua.

摘要: 为构建多孢木霉(Trichoderma polysporum) HZ-31菌株胁迫下野燕麦(Avena fatua L.)基因表达的RT-qPCR技术体系,筛选稳定内参基因,本研究利用GeNorm,NormFinder,BestKeeper和RefFinder 4个程序,对接种多孢木霉HZ-31菌株的野燕麦样本中8种候选内参基因(18S,28S,TUA,UBC,ACT,GAPDH,TBPEF-1α)进行基于SYBR Green的RT-qPCR分析。结果表明TBP,18SUBC是木霉侵染时野燕麦中表达最稳定的内参基因,TBPTUA,TBPGAPDH,18STBP,UBC18S为最适宜两内参基因组合,可作为多孢木霉HZ-31菌株与野燕麦互作过程中功能基因表达分析的内参基因。
