基于三种分子标记的栽培金鸡菊遗传多样性及亲缘关系分析

Abstract: This study aimed to provide a molecular basis for the taxonomic identification,parents traceability and breeding of cultivated Coreopsis. We used ISSR marker,internal transcribed spacer 2 (rDNA-ITS2) sequence,cpDNA psbA-trnH sequence,and cpDNA trnL-F to analyze the genetic diversity and relationship of 100 Coreopsis germplasms widely cultivated around the world. The results showed that:all three molecular markers showed that C. bigelovii was distantly related to other tested materials. 12 ISSR primers amplified a total of 152 bands,and 149 bands were polymorphic (PPB=97.53%,Na=1.9739,Ne=1.4865,H=0.2886,I=0.4406). 100 Coreopsis germplasms could be divided into four groups at a genetic distance of 0.664 based on ISSR marker. The length of ITS2,psbA-trnH and trnL-F sequences we obtained were 171 bp~175 bp,252 bp~426 bp,and 698 bp~703 bp,respectively. 100 Coreopsis germplasms could be divided into six groups based on maternal traceability of psbA-trnH and trnL-F analysis,and their combination defined nine haplotypes. The MP phylogenetic tree also showed that section Calliopsis and section Eublepharis probably could be the female parents to C. verticillata ‘Moonbeam’,C. pubescens ‘Rainbow Lights’ and some Coreopsis cultivars with unique ray floret main colors. Based on NJ phylogenetic tree of ITS2,100 Coreopsis germplasms could be divided into five groups. ITS2 sequences also defined 63 haplotypes,most haplotypes contained only one Coreopsis germplasm. The ITS2 sequence can be used as a DNA barcode for the effective identification of Coreopsis garden cultivars.

摘要: 利用ISSR、核基因ITS2、叶绿体基因psbA-trnHtrnL-F三种分子标记对100个国内外广为种植的栽培金鸡菊(Coreopsis)种质进行遗传多样性和亲缘关系分析,以期为栽培金鸡菊的分类鉴定、育种及指纹图谱构建提供更多分子依据。结果表明:三种分子标记均显示沙漠金鸡菊与其余材料亲缘关系较远;12条ISSR引物扩增出总条带153个,多态性条带149个,多态性条带百分比为97.39%,在遗传距离为0.664处整体可划分为4类;ITS2,psbA-trnHtrnL-F序列大小分别为171bp~175bp,252bp~426bp和698bp~703bp;基于psbA-trnHtrnL-F序列的母系溯源整体可分为6类,二者合并后定义了9个单倍型,两色组和歧序组是具有特殊舌状花色的金鸡菊品种重要的母系材料,轮叶金鸡菊‘月光’和短毛金鸡菊‘虹光’等品种源于远缘杂交;ITS2序列将整体划分为5类,定义了63个单倍型,大部分单倍型只对应一个种质,ITS2序列有望作为金鸡菊品种鉴定的DNA条形码。
