紫花苜蓿CNGC基因家族成员鉴定及分析

Abstract: The cyclic nucleotide-gated channels (CNGCs) gene family functions as one of the non-selective cation channel gene families and plays important roles in physiological processes such as plant signal transduction,growth and development,and environmental stress. In this study,we analyzed the evolutionary relationship,gene structure,conserved motifs,cis-acting elements,chromosome localization,collinearity,and gene expression patterns of the CNGC family members in alfalfa using bioinformatic tools and transcriptomic data. The results showed that there were 16 members of MsCNGC genes family in alfalfa,which were unevenly distributed on chromosomes,with segmental repeation,and most of proteins were located in the plasma membrane. The phylogenetic tree divided MsCNGCs into four subfamilies. Gene structure and conserved motif analysis showed that cNMP/CNBD and ITP functional domains were present. Cis-acting element analysis showed that MsCNGC genes contained many elements that responded to abiotic or biological stresses and hormones. There was interaction between MsCNGC4 and MsCNGC7.2 proteins. MsCNGC genes had tissue specificity. MsCNGC genes had significant response to drought,salt and hormone stress,which provided a theoretical basis for further research on the potential function of MsCNGC genes in the regulation of abiotic stress.

摘要: 环核苷酸门控通道(CNGC)基因家族作为非选择性阳离子通道基因家族之一,在植物信号转导、生长发育和环境胁迫等生理过程中发挥着重要作用。本研究利用生物信息学手段和转录组数据对紫花苜蓿CNGC家族成员的进化关系、基因结构、保守基序、顺式作用元件、染色体定位、共线性关系以及基因表达进行分析。结果表明,紫花苜蓿MsCNGC基因家族成员有16个,在染色体上不均匀分布,存在片段重复,大多数蛋白定位于细胞质膜。系统发育树将MsCNGCs为4个亚家族。基因结构和保守基序分析表明,都含有cNMP/CNBD和ITP功能域。顺式作用元件分析表明,MsCNGCs基因含有许多与非生物或生物胁迫和激素响应元件。MsCNGC4和MsCNGC7.2蛋白之间存在互作。MsCNGC基因具有组织表达特异性。MsCNGC基因对干旱、盐和激素等非生物胁迫均有显著的响应,为进一步研究MsCNGC基因在调控非生物胁迫过程中的潜在功能提供了理论依据。
