响应面法优化火绒草中总黄酮与总酚酸的提取工艺研究

Abstract: In order to explore the optimal extraction process and antioxidant activity of total flavonoids and total phenolic acids in wild Leontopodium leontopodioides,the effects of extraction temperature,cycle times,heating time and extraction pressure on the extraction content of total flavonoids and total phenolic acids in Leontopodium leontopodioides were explored by rapid solvent extraction,and the optimal extraction process of active substances was determined by the single-factor experiment and the response surface design. The results showed that the optimal extraction process for the extraction of total flavonoids in Leontopodium leontopodioides using 70% ethanol was 93℃extraction temperature,3 times,20 min extraction time,and 102 bar extraction pressure. Extraction content was (45.45±0.36) mg·g-1.The best extraction process of total phenolic acid was 90℃extraction temperature,3 times,19 min extraction,and 97 bar extraction pressure. Extraction content was (23.52±0.34) mg·g-1.The IC50 of Leontopodium leontopodioides total flavonoid extract against DPPH radicals and ABTS radicals were 0.012 mg·mL-1 and 0.356 mg·mL-1,respectively. It had a certain reducing power. This study provided a reliable theoretical basis and detailed scientific reference for the extraction process and processing and utilization of the active substance of Leontopodium leontopodioides.

摘要: 为探究野生火绒草(Leontopodium leontopodioides(Wild.)Beauv)总黄酮与总酚酸最佳提取工艺及抗氧化活性,本试验采用快速溶剂萃取法探索萃取温度、循环次数、加热时间及萃取压力对火绒草总黄酮、总酚酸提取含量的影响,通过单因素试验和响应面设计法确定了活性物质的最佳提取工艺。结果表明:当使用70%乙醇提取火绒草中总黄酮时最佳提取工艺为:萃取温度93℃,循环次数3次,加热时间20 min,萃取压力102 bar,提取含量为(45.45±0.36) mg·g-1;总酚酸最佳提取工艺为:萃取温度90℃,循环次数3次,加热时间19 min,萃取压力97 bar,提取含量为(23.52±0.34) mg·g-1。火绒草总黄酮提取液对DPPH自由基和ABTS自由基的IC50为0.012 mg·mL-1和0.356 mg·mL-1,具有一定的还原力。本研究可为火绒草活性物质的提取工艺及加工利用提供可靠的理论依据和详实的科学参考。
