西北不同地理种群红色型豌豆蚜的遗传多样性

Abstract: The genetic structure and genetic diversity of Acrythosiphon pisum (Harris)(Pink form) among five geo-populations from Northwest China were analyzed using simple sequence repeat (SSR). Results showed that 46 polymorphic loci were detected with 15 SSR primers, and the percentage of polymorphic bands (PPB) was 100%. The average number of alleles per locus was 1.7391, while the effective number of alleles was 1.3338. The gene diversity indices (Nei's) were ranged from 0.1448 to 0.3132, and the population from Xinjiang had the highest diversity value. Shannon information index (I) and the percentage in polymorphic loci were 0.3256 and 73.91%. The genetic diversity of population genetic differentiation coefficient (Gst) was 0.3198, indicating the degree of genetic differentiation among populations was greater. In addition, AMOVA showed that the variation among populations accounted for 39%, while the change within populations was 61%. The cluster analysis indicated that the populations from Guyuan, Minhe, Lanzhou and Linxia clustered into one group, and the Hutubi population clustered alone group with the similarity coefficient of 0.56.

摘要: 为探讨西北地区红色型豌豆蚜(Acyrthosiphon pisum (Harris)(Pink form))不同地理种群的遗传结构,了解其遗传多样性,采用微卫星技术(simple sequence repeat, SSR),用15对引物,对5个地理种群进行了测定和聚类分析。结果表明:共检测出46个多态性位点、多态性条带百分率(PPB)为100%、等位基因数1.7391、有效等位基因数1.3338。基因多样性指数(Nei's)在0.1488~0.3132之间,其中新疆呼图壁种群的多样性最高。Shannon信息指数(I)为0.3256、多态位点百分率73.91%。种群基因分化系数Gst为0.3198,表明种群间遗传分化程度较大。AMOVA分析表明,种群间变异占39%,种群内变异占61%。聚类分析表明在相似系数为0.56时,固原、民和、兰州、临夏的种群聚为一类,呼图壁种群独自聚为一类。
