柳枝稷基因枪转化法中国产裂解膜代替进口膜的可行性研究

Abstract:

Comparing domestic rupture disk with imported one in the particle bombardment of switchgrass calluses, the burst pressure of rupture disk, the rate of hyg-resistance callus, the number of regeneration plants and the rate of transgenic plants were analyzed. The results showed that the stability of domestic rupture disk was poorer than that of imported one, whereas the average pressures of both disks were not significant difference. All tested indexes except the rate of resistant callus were also no significant difference between domestic rupture disks and imported ones. In conclusion, the domestic rupture disk could completely replace imported and these results could provide a theoretical premise to save scientific research funds in the particle bombardment.

摘要:

利用国产和进口裂解膜转化柳枝稷(Panicum virgatum L.)愈伤组织,对裂解膜裂解时的压强、抗性愈伤率、分化再生苗及阳性转化率进行统计分析。结果表明:国产裂解膜在裂解时压强的稳定性略低于进口膜,但其平均值差异不显著;除抗性愈伤率国产裂解膜显著低于进口膜外(P<0.05),其余各指标差异均不显著。因此,在柳枝稷基因枪转化法中国产裂解膜完全可以代替进口膜,节约试验经费。
