溶磷红豆草根瘤菌的分离和筛选

Abstract:

In order to obtain phosphate solubilizing Rhizobium strains of sainfoin (Onobrychis viciaefolia). Winogradsky N-free medium and PKO medium were used to isolate and screen Rhizobium strains from sainfoin root nodules. By analyzing the capabilities of sparingly inorganic phosphorus solubilizing, IAA secreting and the nodule nitrogenase activity of screened strains, three rhizobia strains RS-1, RS-8 and RS-14 were obtained. And the phosphate solubilizing capability of RS-1 strain reached 6.98 mg·L-1·d-1 in 14 days, and RS-1 with higher IAA secreting capability and could survive at pH values ranging from 4 to 12,and salt tolerance capability at 11%. Results showed that the screening method used was simple and effective, and the isolated strains could have good prospect of application.

摘要:

为获得溶磷红豆草(Onobrychis viciae folia)根瘤菌(Rhizobium sp.),本研究以Winogradsky无氮培养基和PKO溶磷培养基从红豆草根瘤中分离菌株。按解磷-固氮-结瘤的顺序逐级筛选,结合培养观察和回接鉴定。最终获得3株溶磷根瘤菌RS-1,RS-8和RS-14。其中RS-1产生长素能力及回接根瘤固氮酶活性均较高,溶磷量6.98mg·L-1·d-1,适应pH4~12的环境并耐受11%的盐胁迫。表明该筛选方法简便有效,菌株应用前景好。
