Genetic Diversity of Medicago varia Martyn.Germplasms Detected by SRAP Markers

杂花苜蓿种质SRAP标记遗传多样性研究

Abstract: SRAP(sequence-related amplified polymorphism) molecular markers were used to detect the genetic diversity of 31 accessions of Medicago varia Martyn.from China(Xinjiang),Kazakhstan,and Poland,etc.in May 2009.The objectives of this research were to explore the genetic variation and relationship among M.varia accessions from Central Asia,Europe,and other regions and provide a theoretical basis for its breeding and rational utilization.The results show: a total of 179 bands were detected in 17 pairs of primer combinations,8.41 polymorphic bands in average were amplified for each pair of primers,and the percentage of polymorphic bands(PPB) was 79.89%.The Nei′s genetic similarity coefficient of the tested accessions ranged from 0.59 to 0.874 and the average was 0.723.These results suggest that there is a rich genetic diversity with the Xinjiang resources in comparison with those from Central Asia and Europe.UPGMA cluster analysis indicates the tested materials could be divided into 3 groups,and the materials from China(Xinjiang) were gathered in the first group and third group;they were rich in the genetic variation and had a close phylogenetic relationship with those from Central Asia.Moreover,the phylogenetic relationship among different M.varia accessions was closely related to the geographic environment.

摘要: 利用SRAP(Sequence-related Amplified Polymorphism)分子标记技术,于2009年5月对来自中国新疆、哈萨克斯坦、波兰等地区的31份杂花苜蓿(Medicago varia Martyn.)种质进行遗传多样性研究,旨在探究中亚及欧洲等地区杂花苜蓿种质的遗传变异和亲缘关系,为杂花苜蓿品种选育和利用提供参考。结果表明:17对引物组合共检测出179条清晰条带,平均每对引物扩增出8.41条具有多态性的条带,多态性条带比率(PPB)为79.89%,材料间的相似性系数在0.59到0.874之间,平均相似性系数值为0.723,与中亚和欧洲的参试种质材料相比,新疆的材料具有较为丰富的遗传多样性;UPGMA聚类结果显示,可将供试材料分为3类,来自中国新疆的7份材料分别聚在第1类和第3类,反映出中国新疆的供试材料间遗传变异丰富,与中亚材料有着密切的亲缘关系;不同杂花苜蓿材料的亲缘关系与地理环境具有密切的联系。
