胶质芽孢杆菌遗传多样性ISSR分析

Abstract: The Bacillus mucilaginosus was widely used for the production of microbial fertilizer by its ability to degrade soluble soil mineral compounds with the release of potassium and water-soluble phosphorous.The objective of this study was to assess the genetic diversity of Bacillus mucilaginosus isolated from rhizosphere of different gramineous plant.The genetic variation of 31 Bacillus mucilaginosus accessions has been analyzed by Inter-simple sequence repeats(ISSR) markers.Eighteen screened from 100 general-purpose primers generated 112 highly repeatable polymorphic bands.A dendrogram constructed on the basis of the unweighted pair group method arithmetic average(UPGMA) clustering algorithm revealed that 31 accessions formed two primary distinct clusters.Genetic similarity coefficients calculated from the ISSR data ranged from 0.21 to 0.97 with the lowest value of 0.21 measured between K04 and k17 from Taigu,Shanxi and Sanya,Hainan,K08 and K25 from Beijing and Wenchang,Hainan,respectively.The highest value of 0.97 was measured between K03 and K04 from Taigu,Shanxi.The cophenetic correlation coefficient(r) was 0.94,indicative of a very good fit between the data matrix and the resulting cluster analysis.Principal coordinate analysis(PCO) clearly grouped 31 accessions on the three axes,and the PCO clustering pattern corresponded well with the dendrogram(GS=0.55).In conclusion,these results revealed a high genetic diversity among Bacillus mucilaginosus accessions isolated from various plant rhizosphere,and the clustering and geographical origin of accessions were correlated obviously.

摘要: 胶质芽孢杆菌(Bacillus mucilaginosus)具有分解土壤矿物释放钾素和水溶性磷的能力而被广泛用于微生物肥料的生产,为了解不同禾本科植物根际胶质芽孢杆菌遗传多样性,利用ISSR标记对31份胶质芽孢杆菌材料进行遗传差异分析。从100条通用ISSR引物中筛选到18条重复性好、条带清晰的引物,31份材料共产生多态性条带112条,依据ISSR数据,利用非加权平均法(UPGMA)构建聚类树,将31份胶质芽孢杆菌材料分为主要的2个聚类;材料间的遗传相似系数值为0.21-0.97;其中菌株K04(山西太谷)与K17(海南三亚)以及K08(北京)与K25(海南文昌)之间的遗传相似系数最小(0.21),从山西太谷不同植物根际分离的菌株K03与K04之间的遗传相似系数最大(0.97);协表相关系数(r)为0.94,表明聚类分析的结果与原遗传距离矩阵之间的拟合程度极高。利用主成分分析可将31个材料清晰地分为3个类群,与聚类树(GS=0.55)所得到的结论相一致。研究结果表明不同植物根际分离的胶质芽孢杆菌材料间表现出较为丰富的遗传多样性,同时表明菌株的聚类结果和其地域性分布之间呈现出较明显相关性。
