SRAP分子标记在假俭草杂交后代真实性鉴定中的应用

Abstract: The hybrids status from two hybridization combinations of Eremochloa ophiuroides(Munro)Hack with four parents(E022,E142,E102,E092(1))were tested using SRAP primer combinations with clear and stabilized paternal characteristic bands selected from 400 primer combinations.The results show:(1)51 primer combinations with paternal characteristic bands were selected in A(E102×E092(1))and 87 primer combinations were selected in B(E142×E022).(2)Six primer combinations(303,297,283,180,332 and 358)were used to identify the hybrids status in A(E102×E092(1))and of which,89 were true hybrids.Four primer combinations(18,199,221 and 288)were used to identify the hybrids status in B(E142×E022)and of which,83 were true hybrids.These true hybrids could be useful in the genetic linkage map construction and QTL mapping of important characters.

摘要: 以假俭草(Eremochloa ophiuroides(Munro)Hack)两个杂交组合的4个亲本(E022、E142、E102、E092(1))及其杂交后代为材料,从400对SRAP引物组合中筛选出具有父本特征带的引物,选取其中带型稳定、清晰的引物组合进行杂种真实性鉴定。结果表明:(1)A(E102×E092(1))组合的两个亲本间具有父本特征带的引物组合共有51对,B(E142×E022)组合的两个亲本间具有父本特征带的引物组合共有87对。(2)应用6对SRAP引物(303、297、283、180、332和358)对A(E102×E092(1))组合进行杂种真实性鉴定,其中真杂种数目为89个;应用4对SRAP引物(18、199、221和288)对B(E142×E022)组合的杂交后代进行真实性鉴定,其中真杂种的数目是83个。上述鉴定的假俭草真杂种可以用于构建假俭草遗传连锁图谱及重要性状的QTL定位等研究。
