Abstract

Abstract: To overcome the limitations of an imperfect mutagenesis breeding system for Leymus chinensis and the shortage of germplasm with high seed setting rates， this study established an efficient mutagenesis technology using a combination of spatial and atmospheric pressure room-temperature plasma （SP+ARTP）. A high-throughput， accurate phenotypic analysis of large-scale mutant populations was achieved by utilizing an automatic seed setting rate identification model based on the YOLO algorithm. The results indicated that cultivar ‘Xiwuzhumuqin’ was the most suitable material for mutagenesis； a 60-minute treatment with 20% NaOH was the optimal pretreatment method. Single-space mutagenesis significantly promoted seed germination and seedling growth. The mutagenesis efficiency of the M₁ generation using the SP+ARTP combination reached 1.20%， and 23 new germplasm lines with high seed setting rate were successfully selected. These lines exhibited a seed setting rate of 30% higher than that of the control group. The combined mutagenesis and high-throughput phenotyping system developed in this study provides key technical support and germplasm resources for the targeted breeding of high-seed-setting L. chinensis varieties and their industrial application.

摘要： 为突破羊草（Leymus chinensis）诱变育种体系不完善与高结实率种质缺乏的瓶颈，本研究建立了空间与常压室温等离子体复合（SP+ARTP）的高效诱变技术体系。利用基于YOLO算法构建的结实率自动识别模型，实现了对大规模诱变群体表型的高通量、精准鉴定。结果表明，‘西乌珠穆沁’为最适诱变受体，20% NaOH处理60分钟为最佳前处理方法；单一空间诱变可显著促进种子萌发与幼苗生长；SP+ARTP复合诱变的M₁代诱变效率达1.20%，并成功筛选出23份高结实率新种质，其结实率较对照均提升30%以上。该研究所建立的复合诱变与高通量表型鉴定体系，为羊草高结实率品种的定向选育及产业化应用提供了关键技术支撑与种质资源。
