摘要

大型真菌主要为担子菌门的真菌和少数为子囊菌门的真菌,该类真菌具有重要的经济价值和生态功能,主要生长在森林生态系统中。30年来作者对我国几乎所有类型森林生态中的大型真菌进行了系统调查和采集,共采集标本11.2万号。基于对这些材料的形态学及分子系统学研究,并结合生态学和生物地理学特征,共鉴定出中国森林大型真菌4 250种,隶属于担子菌门和子囊菌门的21个目,发现和发表2个新科、4个新亚科、69个新属和885个新种。云南省是我国森林大型真菌最丰富的省份,描述于该省的新种有314种,占作者发表的全部中国新种的35%。这些研究为深入认识全球大型真菌物种多样性提供了中国的贡献,更新了我国重要食药用菌名称,揭示了我国毒蘑菇多样性基本特征,系统论述了我国森林病原菌的物种多样性,为资源利用、森林健康和保护提供了科学依据;论述了森林大型真菌代表性类群在种和属级水平的起源和演化,为今后开展重要类群科级、目级甚至纲级的系统进化关系提供了重要数据。

Abstract

Macrofungi, as an important component in forest ecosystems, consist of most members of Basidiomycota and some members of Ascomycota, having important economical value and ecological functions. Extensive field investigations have been carried out in almost whole types of the Chinese forests during the past 30 years, and 112 000 specimens were collected. Based on morphological examination and phylogenetic analyses in combination with ecology and biogeography, 4 250 species belonging to 21 orders in Baidiomycota and Ascomycota were identified, including two new families, four new subfamilies, 69 new genera and 885 new species. Yunnan Province is the richest in macrofungal diversity among provinces or regions in China, and 314 new species were described from this province, accounting for 35% of all the new species described from China by the authors. Our studies have made contributions to deepening the understanding of global diversity of macrofungi. The names of some important Chinese medicinal fungi were revised, the diversity characteristics of Chinese poisonous mushrooms were revealed, and the pathogenetic wood-decaying species were ascertained. These data improved our knowledge on utilization of natural resources and protection of forest health. Based on molecular evidences, the origin of some forest representative fungal genera or species complex were deduced, and their dispersal and speciation were discussed, for the purposes of providing some data for evolutionary study at level of family, order or class of macrofungi henceforth.

大型真菌通常是指肉眼可见、徒手可摘的真菌,主要为担子菌门Basidiomycota、少数为子囊菌门Ascomycota真菌。该类真菌具有重要的经济价值:目前绝大多数野生或栽培的食用菌,包括松茸Tricholoma matsutake (S. Ito & S. Imai) Singer、香菇Lentinula edodes (Berk.) Pegler和黑木耳Auricularia heimuer F. Wu et al.等都是大型真菌(戴玉成等 2010);同样我国绝大多数药用真菌,包括灵芝Ganoderma lingzhi Sheng H. Wu et al.和猴头Hericium erinaceus (Bull.) Pers.等也都属于大型真菌(戴玉成和杨祝良 2008)。该类真菌还具有重要的生态价值:最主要的外生菌根真菌如牛肝菌科Boletaceae和红菇科Russulaceae,最重要的木材降解菌如多孔菌科Polyporaceae和锈革孔菌科Hymenochaetaceae等科的种类也都是大型真菌(李玉等 2015)。该类真菌也影响森林健康:一些重要的森林病原真菌,包括异担子菌属Heterobasidion Bref.和蜜环菌属Armillaria (Fr.) Staude等的种类能够造成重大森林病害(戴玉成 2012)。该类真菌还影响人类健康:一些种类是剧毒蘑菇,据国家食品安全风险评估中心统计,2020年我国有约8 000人误食毒蘑菇,74人死亡,准确鉴定这些剧毒蘑菇对预防和治疗蘑菇中毒具有重大意义。因此,大型真菌在生态系统中对促进植物生长、维护生态系统的稳定性及物质循环和能量流动发挥不可替代的作用,也在一定程度上影响森林和人类的健康。

大型真菌在我国和世界其他地区都是最早被人类认识和利用的生物类群之一,在8 500年前宁夏银川境内贺兰山上就有关于灵芝的岩画(李玉等 2015),在6 800年前新石器时期的浙江地区已有使用灵芝的遗迹(袁媛等 2018)。约在2 000年前的《神农本草》中就有猪苓多孔菌Polyporus umbellatus (Pers.) Fr.等大型真菌的记载(无名氏 102-200)。

在用双名法最早描述的植物中,就包括了木蹄层孔菌Fomes fomentarius (L.) Fr. ≡ Boletus fomentarius L.、火木层孔菌Phellinus igniarius (L.) Quél. ≡ Boletus igniarius L.和点柄乳牛肝菌Suillus granulatus (L.) Roussel ≡ Boletus granulatus L.等大型真菌(Linnaeus 1753)。虽然我国从约2 000年前就开始对大型真菌进行了诸多的记载和药用功能描述,但以双名法规对我国大型真菌进行分类学研究始于18世纪。一些外国传教士在中国进行植物标本采集时也一并采集了大型真菌,并进行了系列报道。最早以我国材料记载的大型真菌为1782年发表的五棱散尾鬼笔Lysurus mokusin (L.) Fr. ≡ Phallus mokusin L.,其标本是由法国传教士西博特(P.M. Cibot)在中国采集的(戴芳澜 1979a)。我国著名真菌学家邓叔群院士是中国大型真菌研究的先驱,他在20世纪30年代对我国大型真菌进行了系列报道(Teng 1934,1935,1936,1939)。1979年出版的《中国真菌总汇》中记载了大型真菌种级分类单元名称2 020个,其中担子菌1 820种(戴芳澜 1979b)。

近40年来诸多省份出版了地方大型真菌志,但全国范围内具有重要影响的大型真菌专著有卯晓岚(2000)出版的《中国大型真菌》和李玉等(2015)出版的《中国大型菌物资源图鉴》。前者记载了1 701种,后者报道了1 819种。这2部专著均对我国已知的主要大型真菌进行了描述和记载并附有照片,对促进我国大型真菌的研究和开发具有重要指导意义。

尽管我国科学家对中国大型真菌资源进行了系列研究,但若与植物多样性研究相比,大型真菌多样性研究仍然严重滞后,物种多样性认识严重不足,诸多系统学问题有待解决。特别是由于大型真菌化石较少,多数类群的起源和演化问题处于探索阶段。在各种生态系统中,森林生态系统植被区系组成复杂、生境多样,孕育了丰富的大型真菌资源。因此森林生态系统的大型真菌物种多样性最高,但过去的研究还不够充分和深入。北京林业大学、中国科学院昆明植物研究所和中国科学院沈阳应用生态研究所的学者30年来对全国所有省区市550多个国家公园、自然保护区、森林公园和林场的森林大型真菌多样性和资源进行了广泛考察,获得标本近11.2万号,保藏于上述3个单位的标本馆中(BJFC、KUN和IFP)。基于对上述材料的系统学研究,我们取得的主要研究结果总结如下。

1 发现中国森林大型真菌4 250种

基于标本的形态学、生态学或分子系统学研究数据,共鉴定出4 250种中国森林大型真菌,隶属于担子菌门的伞菌目Agaricales、淀粉孢革菌目Amylocorticiales、阿太菌目Atheliales、木耳目Auriculariales、牛肝菌目Boletales、鸡油菌目Cantharellales、伏革菌目Corticiales、花耳目Dacrymycetales、地星目Geastrales、褐褶菌目Gloeophyllales、钉菇目Gomphales、锈革孔菌目Hymenochaetales、鬼笔目Phallales、多孔菌目Polyporales、红菇目Russulales、蜡壳菌目Sebacinales、革菌目Thelephorales、糙孢菌目Trechisporales和银耳目Tremellales,以及子囊菌门的肉座菌目Hypocreales和盘菌目Pezizales等21个目中(Yang 1997;杨祝良 2005,2015;Dai 2011,2012;Wu et al. 2014,2016;Cai et al. 2016;杨祝良等 2017,2019;Cui et al. 2018,2019;戴玉成 2018;Han et al. 2019;Li et al. 2020)。

2 发现森林大型真菌重要类群新分类单元960个

利用形态特征、分子数据和生态习性等多相证据,发现和发表2个新科、4个新亚科、69个新属和885个新种,具体情况如下。

3 新种涉及高等真菌各大主要纲目

研究发现的885个新种隶属于真菌界的2个门(子囊菌门和担子菌门)、5个纲(盘菌纲Pezizomycetes、粪壳菌纲Sordariomycetes、伞菌纲Agaricomycetes、花耳纲Dacrymycetes和银耳纲Tremellomycetes)、17个目、63个科、247个属,另有5个新种的科级分类位置不定。其中担子菌门种类861种,占97.3%;伞菌纲种类有859种,占97%,因此绝大多数新种为担子菌门伞菌纲的种类。885个新种在目级分类单元的分布见图1,其中多孔菌目、锈革孔菌目、伞菌目、牛肝菌目、革菌目和红菇目分别有280、187、156、99、55和54种,分别占新种总数的31.6%、21.1%、17.6%、11.2%、6.2%和6.1%,这6个目包括的新种占全部新种的94%。在科级水平上锈革孔菌科、多孔菌科、牛肝菌科、鹅膏菌科Amanitaceae、革菌科Thelephoraceae和齿耳科Steccherinaceae分别有163、131、95、84、52和31种,分别占新种总数的18%、11%、10%、9%、6%和4%,这6个科包括的新种占全部新种的63%。上述数据表明在这些目和科中我国的大型真菌多样性较为丰富,研究也较为深入。

4 新种主模式产地覆盖全国30个省区

上述885个新种及模式标本产地在全国的分布见图2。从图2可以看出描述自云南省的新种最多,有314种,占全部新种的35%,是全国最丰富的省份;其次是海南省有110种,占12%;西南地区(云南、西藏、四川、贵州和重庆)的新种有404种,占全部新种的46%,是最丰富的地区,其次是华南地区(海南、广东、广西和湖南)有219种,占25%。

5 我国重要食药用真菌名称得到及时更新

我国最著名的药用真菌灵芝从1907年(Patouillard 1907)被鉴定为欧洲灵芝Ganoderma lucidum (Curtis) P. Karst.后,一直到2012年才被证实它与欧洲灵芝不同,为一个新种,并以灵芝的汉语拼音命名为G. lingzhiCao et al. 2012)。无独有偶,在19世纪后期Kalchbrenner & Thümen(1881)将我国著名的食用菌黑木耳鉴定为欧洲木耳 Auricularia auricula (L.) Underw = A. auricula-judae (Bull.) Quél.,一直到2014年才被证实中国的黑木耳与欧洲木耳是两个不同的物种,故黑木耳被命名为A. heimuerWu et al. 2014)。研究还发现,在东亚地区广泛栽培的冬菇(俗称“金针菇”)过去一直认为是欧洲毛腿冬菇Flammulina velutipes (Curtis) Singer,但实际上东亚的冬菇和欧洲的毛腿冬菇是不同的物种,最近才被更新为F. filiformis (Z.W. Ge, X.B. Liu & Zhu L. Yang) P.M. Wang, Y.C. Dai, E. Horak & Zhu L. Yang(戴玉成和杨祝良2018;Wang et al. 2018)。此外,对我国药用真菌物种多样性进行全面更新,中国药用真菌有540种,最主要的药用功能为增强免疫力、抗肿瘤、抗氧化和抗细菌(戴玉成和杨祝良 2008;Dai et al. 2009)。

6 毒蘑菇研究成果支撑全国毒蘑菇中毒防治工作

2000年3月,9名务工人员在广州误食野生白色蘑菇,导致8人中毒死亡,经过研究发现该毒蘑菇是个新种,并命名为致命鹅膏Amanita exitialisYang & Li 2001)。致命鹅膏在我国至少引发了30余起中毒事件,造成100余人中毒,其中超过60人死亡,是我国最著名的剧毒蘑菇之一(陈作红等 2016)。此外,研究还发现了拟灰花纹鹅膏Amanita fuligineoidesZhang et al. 2010)、灰盖粉褶鹅膏Amanita griseoroseaCai et al. 2016)、浅红鹅膏Amanita pallidoroseaZhang et al. 2010)、裂皮鹅膏Amanita rimosaZhang et al. 2010)、毒环柄菇Lepiota venenata、毒沟褶菌Trogia venenata等剧毒蘑菇。在我国的大型真菌中,共有毒蘑菇480种,对子实体发生时间和地理分布特征有了较为深入的认识(Wu et al. 2019),为毒蘑菇准确鉴定提供了重要依据,对预防和治疗蘑菇中毒具有重要现实意义。

7 我国森林病原真菌物种多样性得到系统论述

研究过程中共发现造成我国木本植物腐朽病害的病原菌152种(戴玉成 2012),其中栎生多孢孔菌Abundisporus quercicola Y.C. Dai和罗汉松邦氏孔菌Bondarzewia podocarpi Y.C. Dai & B.K. Cui等49种(占总数的34%)为研究过程中在我国首次报道为林木病原菌。对每个病原菌的寄主、侵染部位、腐朽类型、发生频率及分布进行了报道。135种(占总数的89%)造成木材白色腐朽,17种(占总数的11%)造成褐色腐朽。对世界范围内最严重森林病害即针叶树干基腐朽病的病原菌进行了系统研究,在我国针叶树干基腐朽病的病原菌过去普遍被认为是多年异担子菌Heterobasidion annosum (Fr.) Bref.,该真菌最初发现于欧洲,过去基于传统的分类鉴定方法认为该病原菌在我国东北和西南等地区以及国外的美洲地区也广泛分布(袁嗣令 1997)。最新的研究发现我国广泛分布的异担子菌种类实际上是亚小孔异担子菌H. subparviporum,而真正的多年异担子菌在我国并没有分布(Yuan et al. 2021)。世界范围内能够引起针叶树干基腐朽病的病原菌包括欧洲的冷杉异担子菌H. abietinum Niemelä & Korhonen、小孔异担子菌H. parviporum Niemelä & Korhonen和多年异担子菌及美洲的西方异担子菌H. occidentale Otrosina & Garbel.和变孔异担子菌H. irregulare Garbel. & Otrosina,这几种造成针叶树干基腐朽病严重危害的种类在我国并没有分布,应将这些种类列为重要入境检疫对象(Yuan et al. 2021)。

8 森林大型真菌代表性类群的起源和演化规律始有认识

异担子菌属Heterobasidion种类引起世界范围内针叶树最严重的森林病害。基于多基因片段数据,运用最大似然法和贝叶斯法对该属的12个种进行了系统发育关系分析,发现异担子菌属可能起源于中新世早期的欧亚大陆,随后在中新世中期和上新世早期扩散至其他大陆并形成物种。变孔异担子菌和西方异担子菌分别越过白令海峡和北大西洋路桥在北美成种,南半球南洋杉异担子菌H. araucariae P.K. Buchanan可能是由于最近人类迁徙引入的。板块构造和长距离扩散是影响异担子菌成种和生物地理分布的最可能因素(Chen et al. 2015)。

檐托鹅膏组Amanita sect. Phalloideae (Fr.) Quél.是鹅膏属中较为独特的一个类群,因其含有致命的环肽类毒素而闻名。综合形态学、解剖学和生态学特征及相关生物化学分析研究,作者构建了全球剧毒鹅膏较为全面的分子系统发育框架,共界定了剧毒鹅膏28个系统发育种,研究表明剧这些剧毒鹅膏是一个自然的单系类群,鹅膏毒肽(amatoxins)和鬼笔毒肽(phallotoxins)为单次起源,是剧毒鹅膏的一个共衍征。研究显示剧毒鹅膏起源于古新世早期的古热带地区,在渐新世中期至中新世期间通过白令陆桥传播至北美,后由于中新世后期全球气候变冷,白令陆桥的阻断而发生异域成种事件。扩散和隔离分化在剧毒鹅膏的洲际间断分布格局形成过程中起了重要作用,东亚为剧毒鹅膏现代分布中心和物种多样性中心(Cai et al. 2014)。

松塔牛肝菌属Strobilomyces Berk.真菌为世界广布的外生菌根菌,在全球广泛取样的基础上,通过多基因片段构建了该属较为全面的分子系统发育框架。基于分子钟推算、祖先分布区和宿主植物重建,研究了该属的生物地理,发现该属约在始新世起源于非洲,之后借助“北方热带森林”扩散至东南亚,形成了非洲-东南亚古热带间断分布格局。在渐新世期间从东南亚向北别扩散至东亚、向南扩散至大洋洲。中新世期间通过白令陆桥和中国东海陆桥迁移以及气候变化驱动的隔离分化等在北半球形成间断分布格局。在中新世中期,东亚发生了一次快速辐射演化事件,致使其成为该属的现代分布中心和物种多样性中心(Han et al. 2017)。

糙皮侧耳复合群Pleurotus ostreatus (Jacq.) P. Kumm. complex是具有重要经济和生态意义的腐生真菌。基于全球样品和40个核单拷贝同源基因,运用最大似然法和贝叶斯法对该类群进行系统发育关系分析,共界定了糙皮侧耳复合群20个系统发育种。分子钟分析数据表明,侧耳属的分化可能发生在侏罗纪晚期,而糙皮侧耳复合群的共同祖先可能在始新世末(约39Mya)出现于东亚。在晚渐新世、晚中新世和晚上新世期间糙皮侧耳的种类可能通过北大西洋大陆桥或白令海峡大陆桥分别从东亚迁移至北美,然后通过多次分散和分隔事件在新旧世界同时分化。晚中新世和第四纪冰期循环,干旱加剧和气候变冷事件对北半球物种多样性分化产生了深刻影响(Li et al. 2020)。

9 初步揭示了我国森林大型真菌分布格局

深入分析和系统总结前人研究我国大型真菌及其他地区真菌重要论著成果后,发现我国现在大型真菌区系具有复杂而独特的性质,提出了我国大型真菌呈现出“东喜马拉雅-横断山区”、“华中地区”和“中国热带地区”三大物种多样性中心的分布格局,论证了该格局应是地质历史事件、地理因素、多样化地理成分、新老种汇聚、与植物协同进化和复杂的生态环境等多种因子共同作用的结果。复杂性主要表现为其与世界其他区系如非洲、北美洲、大洋洲有各种联系,融入了多种地理起源迥异的成分;该区系中还汇聚了古老种、孑遗种和新生种等成分。独特性主要表现为该区系3个中心分别具有各自独特的真菌属和种(Yang 2005)。

10 研究展望

研究人员虽然取得了上述研究成果,但尚有许多类群还有待进一步研究。一方面,除了森林生态系统外,还有草原生态系统、荒漠生态系统及农田生态系统的大型真菌资源和多样性研究需要进一步展开。另一方面,今后研究中还需要聚焦在以下几个重点领域:(1)加强薄弱类群和区域大型真菌的深入和系统研究;(2)加强大型真菌DNA条形码研究,以便非专业人士快速鉴别大型真菌物种;(3)对重要类群开展国际合作研究,强强联合,不断提高研究水平;(4)从系统发育基因组角度,研究重要类群的科、目甚至纲级的系统进化关系。

深入认识我国乃至东亚和东南亚的大型真菌多样性的起源和演化规律,可为资源开发和利用奠定科学基础,为揭示森林生态系统物种多样性维持机制提供科学支撑。

"Lingzhi" is a mushroom that has been renowned in China for more than 2,000 years because of its claimed medicinal properties plus its symbolic fortune. "Lingzhi" has high economic value mostly as a dietary supplement in the modern market especially in East Asia, and its medicinal functions have become a hot study topic. For over a century, the highly prized medicinal fungus, known as "Lingzhi" in East Asia, has been assigned to Ganoderma lucidum, a species originally described from Europe. Molecular studies in recent years have revealed that the commercially cultivated 'G. lucidum' ("Lingzhi") in East Asia is a different species from the true G. lucidum. The present study aims to clarify the species identity of "Lingzhi" based on morphological studies and analysis of rDNA nuc-ITS sequences, and additional gene fragments of mt-SSU, RPB1, RPB2, and TEF1-alpha of "Lingzhi" were provided. All Ganoderma species that mostly resemble "Lingzhi" in phylogeny and /or morphology were included for analysis. We propose a new species G. lingzhi for "Lingzhi", which has an East Asia distribution. The most striking characteristics which differentiate G. lingzhi from G. lucidum are the presence of melanoid bands in the context, a yellow pore surface and thick dissepiments (80-120 mu m) at maturity. G. curtisii is most closely related to G. lingzhi in phylogeny and is from North America. Ganoderma flexipes, G. multipileum, G. sichuanense, G. tropicum and 'G. tsugae', are also closely related with G. lingzhi and are reported from China. These species are compared and discussed. 'Ganoderma tsuage' reported from China is determined as conspecific with G. lucidum, hence the distribution of G. lucidum extends from Europe to northeastern China.
