GA3浸种对露蕊乌头种子破眠及萌发生理特性的影响

Influence of GA3 on Breaking Seed Dormancy and Germination Physiological Characteristics of Aconitum gymnandrum

摘要:

露蕊乌头具有重要的药用价值,但自然条件下其种子的萌发率极低。本试验以露蕊乌头种子为材料,在光照和黑暗条件下,用不同浓度的赤霉素(GA3)溶液浸种,探讨GA3浸种对露蕊乌头种子破眠及萌发生理特性的影响。研究表明:GA3浸种可以提高种子的萌发率,其中1 000 mg·L-1 GA3处理下种子的萌发率达到最大值,光照和黑暗条件下分别为91.66%和95.67%。黑暗比光照条件下露蕊乌头种子的发芽势、发芽指数和活力指数分别提高72.19%、26.58%、32.60%。与对照相比,1 000 mg·L-1 GA3浸种可提高α-淀粉酶,蛋白酶,脂肪酶的活性和可溶性蛋白的含量,降低可溶性糖含量,提高SOD、POD及CAT的活性,可见适宜浓度的GA3浸种可有效促进露蕊乌头种子的萌发。

Abstract:

Aconitum gymnandrum is an important medicinal plant material, but its seed germination rate is extremely low under natural conditions. We used Aconitum mnandrum seeds as materials and soaked them in different concentrations of gibberellin acid (GA3) solution under light or dark conditions, to explore the effect of GA3 seed soaking on the seed dormancy-breaking and germination characteristics. The results showed that seed germination rate could be increased by soaking seeds with different concentrations of GA3. In addition, the germination rate reached its maximum under 1 000 mg·L-1GA3 treatment, which was 91.66% and 95.67% under light and dark conditions, respectively. Compared to light conditions, the germination energy, germination index and vigor index of Aconitum gymnandrum seed in dark conditions increased by 72.19%, 26.58% and 32.60%, respectively. Compared to control, the conditions of 1 000 mg·L-1 GA3 can increase the activities of α-amylase, proteinase, lipase, superoxide dismutase(SOD), peroxidase(POD) catalase(CAT), and soluble protein content, and decrease the content of soluble sugar. These founding showed that the appropriate concentration of GA3 can effectively promote germination of Aconitum gymnandrum.
