蒺藜苜蓿种子休眠机制及其破除方法研究

摘要:

以蒺藜苜蓿(Medicago truncatula)种子为试验材料,通过测定种子生活力、发芽率、吸水率及粗提物的生物活性;并用不同浓度的赤霉素(GA3)浸种后4℃低温层积或同浓度硝普钠(SNP)浸种,揭示蒺藜苜蓿种子休眠原因及破除方法.结果表明:蒺藜苜蓿种子具有生活力;30 d内裸粒种子发芽率为100%;破皮种子的吸水率显著高于完整种子;随着蒺藜苜蓿各部分粗提物浓度的升高,白三叶(Trifolium repens)草种子萌发和幼苗生长显著降低;0.1~0.7 mmo·L-1的GA3浸种后低温层积或同浓度SNP浸种均可以显著提高种子的发芽率,其中以0.1 mmol·L-1 GA3浸种后低温层积或同浓度的SNP处理效果最好.表明蒺藜苜蓿种子的休眠属于综合性休眠,可用GA3浸种后低温层积或SNP浸种来解除其休眠.

Abstract:

Using Medicago truncatula seeds as experimental material, the dormancy mechanism of Medicago truncatula seeds was discussed through the determination of seed viability, germination rate, water absorption rate and biological activity tests of the crude extracts; and the breaking methods of seeds were explored with pre-soaking in different concentrations of GA3 or SNP. Experimental results showed that the staining life force of Medicago truncatula seeds was 55.56%; the germination rate of Medicago truncatula seed removed coat was 100%; water absorption test revealed that Medicago truncatula seed coat hindered embryo absorption to water in a certain extent; with the concentrations of crude extract of Medicago truncatula increasing, the inhibitory effects on the seed germination and seedling growth of Trifolium repens were more obvious. Pre-soaking with 0.1~0.7 mmol·L-1 GA3 or SNP promoted seed germination percentage, germination potential, germination index and activity index, in particular, pre-soaking with 0.1 mmol·L-1 GA3 or SNP broke seed dormancy significantly. In conclusion, Medicago truncatula seeds are featured multiple dormancies; pre-soaking with GA3 or SNP solution can break the dormancy of Medicago truncatula effectively.
