60Co-γ辐照对白三叶愈伤组织的诱变效应

The Mutagenic Effects of 60Co-γ Radiation on White Clover Callus

摘要:

为探讨辐射对白三叶(Trifolium repens)胚性愈伤的诱变效应,以白三叶3个品种不同生长时期(50和70 d)的愈伤组织为材料,进行了60Co-γ不同辐照剂量的诱变处理,并统计其诱变后的分化率。结果表明:随着辐射剂量的增加,同一生长时期不同品种的分化率变化规律不一致,同一品种不同生长时期的诱变愈伤组织的分化率也不一致。通过线性直线回归分析70 d愈伤组织的诱变效应,分别得到各品种的半致死剂量,‘瑞文德’为23.18 Gy,‘海法’为19.59 Gy,‘铺地’为4.38 Gy。初步获得叶片增大和叶片变厚、茎部增粗2个变异单株。

Abstract:

Calli of three white clover varieties at different growth stages (50 d and 70 d) were mutagenized using different 60Co-γ irradiation doses and the differentiation rate of mutagenesis was analyzed. The mutagenic effects of radiation on the embryonic callus of Trifolium repens were investigated in this study to provide evidence for white clover breeding. Experimental results showed that with the increase of radiation dose, the variation of differentiation rate was different among different varieties in the same growth period, and the callus differentiation rate of the same species also differed from different growth stages. The semi-lethal doses of all varieties were obtained by analyzing the mutagenic effect of 70 d callus using linear regression analysis, and ‘Rivendel’ is 23.18 Gy, ‘Haifa’ is 19.59 Gy, ‘Prop’ is 4.38 Gy. Two mutant individuals with large and thick leaves, as well as thickening stems were selected.
