苜蓿SKU5基因的表达调控及其在秋眠中的作用

摘要: 为了探索SKU5基因在紫花苜蓿(Medicago sativa L.)中表达调控规律和在苜蓿秋眠中的作用,本研究拟检测春、夏、秋季和人工条件下不同温度、不同日照长度下秋眠型和非秋眠型苜蓿叶片中SKU5基因mRNA相对表达量,分析同一秋眠型苜蓿品种不同时期SKU5基因mRNA相对表达量差异和不同时期2种秋眠型苜蓿品种间SKU5基因mRNA相对表达量的差异,以及SKU5基因mRNA相对表达量与苜蓿株高、叶面积、日照长度、温度的相关性。结果表明:SKU5基因的表达受日照长度极显著负调控,受温度极显著正调控;2种苜蓿叶片中SKU5基因的表达受日照长度和温度的调控结果不一致;秋眠型苜蓿叶中SKU5基因mRNA相对表达量与株高、叶面积呈显著负相关。因此,随着日照长度的缩短,SKU5基因高表达很可能促进秋眠型苜蓿的秋眠。

Abstract: To explore the expression of SKU5 gene in alfalfa (Medicago sativa L.) leaves and its role in alfalfa fall dormancy,we detected the mRNA relative abundance of SKU5 in leaves of fall dormant alfalfa and non-dormant alfalfa in spring,summer and autumn,and at different temperature or different daylength under artificial conditions. Differences of SKU5 mRNA relative abundance in leaves of two fall dormancy type alfalfa varieties at the same growth stage and the seasonal changes of SKU5 gene expression were analyzed. The correlations among SKU5 mRNA relative abundance,leaf area,plant height,daylength and temperature was analyzed. The results showed that SKU5 gene expression was negatively correlated with daylength and positively correlated with temperature. The response of SKU5 gene expression to daylength and temperature were different between the two alfalfa varieties. SKU5 mRNA relative abundance in leaves of fall dormant alfalfa variety was negatively correlated with plant height and leaf area. It was speculated that the high-expression of SKU5 promoted the fall dormancy of alfalfa with the shortening of daylength.
