植物根际促生菌Bacillus mycoides Gnyt1菌株生物学特性比较研究

Comparative Study on the Biological Characteristics of Plant Rhizosphere Growth-Promoting Bacteria Bacillus mycoides Gnyt1

摘要: 为明确保藏后优良促生菌株的生物学特性,本研究以优良植物根际促生菌Bacillus mycoides Gnyt1菌株为研究材料,以4株优良菌株(Pseudomonas sp.Gm92,Ensifer meliloti 426,Bacillus subtilis 89,Ensifer meliloti 93)为对照材料,测定各菌株保藏后的6个指标,明晰保藏后Bacillus mycoides Gnyt1菌株生物学特性的变化。结果表明,Bacillus mycoides Gnyt1菌株的增殖速度比对照菌株快,菌株生长的最适温度为28℃~34℃,最适pH值6~7,溶磷能力、固氮能力及分泌植物激素能力均显著高于对照菌株(P<0.05)。通过比较菌株保藏前后的生物功能,发现保藏后菌株生物性状无明显变化,菌株溶磷能力、固氮能力和分泌植物激素的能力均较高。因此,保藏两年后的Bacillus mycoides Gnyt1菌株仍可继续作为研究菌株用于深入研究其功能基因和促生机理。

Abstract: In order to provide material guarantee for the study of functional genes of excellent growth-promoting strains,in our study,the excellent plant growth-promoting rhizosphere Bacillus mycoides Gnyt1 was used as the research material,and 4 excellent strains (Pseudomonas sp. Gm92,Ensifer meliloti 426,Bacillus subtilis 89,Ensifer meliloti 93) were used as control materials to determine the preservation of each strain. Six indicators were used to analyze the biological characteristic changesof the Bacillus mycoides Gnyt1 strains after preservation. The results showed that the Bacillus mycoides Gnyt1 strain had a faster proliferation rate than the control strains,the optimum temperature was 28℃~34℃,the optimum pH value was 6~7,and the ability of dissolving phosphorus,fixing nitrogen,and secreting plant hormones was significantly higher than that of the control strains (P<0.05). By comparing the biological functions of the strains between the two years of preservation,it was found that the biological characteristics of the strains had no obvious changes,and the degradation of the strain’s ability to solubilize phosphorus,fix nitrogen,and secrete plant hormones were higher. Therefore,the Bacillus mycoides Gnyt1 strain that had been preserved for two years could still be used as an experimental strain for in-depth study of its functional genes and functional gene families,laying the foundation of experimental materials for the next step of gene function verification.
