ISSR标记的早熟禾遗传多样性分析

摘要: 利用ISSR分子标记技术对早熟禾属(Poa L.)6种共31份种质材料进行遗传多样性分析,以期了解不同早熟禾种质材料之间的遗传差异,为早熟禾资源的保护、品种选育和分子鉴定提供理论依据。结果表明:早熟禾属种质间存在丰富的遗传多样性,6个ISSR引物共扩增出111条清晰谱带,平均多态性比率(PPB)为97.79%,Nei’s基因多样性指数(H)为0.4239,Shannon信息指数(I)为0.6137;31份材料间的遗传相似系数(GS)为0.4146~0.9512;通过UPGMA分子系统聚类法,将31份种质材料分为6个大类群,其中高山早熟禾(Poa alpigena L.)与其他材料间的遗传分化最大,单独聚为一类,7个美国的草地早熟禾(Poa pratensis L.)品种聚为一类,聚类结果呈现出一定的地域性分布规律。本研究探讨了早熟禾属种质间亲缘关系及遗传多样性,为其进一步研究与利用提供了分子水平的依据。

Abstract: The genetic diversity of 31 Poa L.accessions(6 species) was analyzed using ISSR markers,in order to understand genetic differences among Poa L.germplasm and provide reference for germplasm resource protection,breeding and molecular identification.Results indicated that there was abundant genetic diversity in Poa germplasm.Total 111 bands were detected by 6 primers,and the percentage of polymorphic bands(PPB) was 97.79%.Nei’s gene diversity index(H) was 0.4239;the Shannon diversity index(I) was 0.6137.The genetic similarity(GS) among 31 accessions was 0.4146~0.9512.The 31 accessions were classified into 6 major groups based on UPGMA cluster analysis.Poa alpigena L.had the highest genetic differentiation and clustered a unique group.Seven American accessions of Poa pratensis L.were classified into one group.The accessions from same origin were classified into the same group indicating the geographical distribution of genetic diversity.The genetic relationships and diversities among Poa germplasm were also discussed for future research and application of Poa germplasm at the molecular level.
