内生真菌对野大麦种子人工老化处理下的生理影响

Effects of Endophytic Fungi on the Seed Physiology of Hordeum brevisubulatum Under Artificial Ageing Treatment

摘要:

以带Epichloë内生真菌和不带Epichloë内生真菌的野大麦(Hordeum brevisubulatum)种子为试验材料,在58±1℃和100%相对湿度条件下设置0,5,10,15,20,25,30和35 min人工老化时间,研究老化过程中种子的活力变化以及内生真菌对种子的生理影响。结果表明:在人工老化过程中,种子的发芽率、发芽势、胚芽胚根长和活力指数随老化时间增加而降低;浸出液的电导率和可溶性糖含量随老化时间的增加而增加,且与发芽率、发芽势、胚芽长、胚根长和活力指数呈极显著负相关(P<0.01),但被内生真菌侵染的种子其老化生理反应变化明显有所减缓。研究认为,与未被内生真菌侵染的种子相比,内生真菌可促进野大麦的发芽,促进芽长根长的生长,降低膜的损伤,减少浸出液电导率和可溶性糖的含量,从而降低种子的劣变程度,延长种子贮藏时限。

Abstract:

The vitality of Hordeum brevisubulatum seeds was investigated with Epichloë endophyte and without Epichloë endophyte under the conditions of 58±1℃ and 100% relative humidity,and the effects of endophytic fungi on seed physiology were discussed under different artificial ageing times (0,5,10,15,20,25,30 and 35mins). The results indicated that the seed germination percentage,germination energy and vigor index of Hordeum brevisubulatum seed as well as the plumule length and root length of seedling decreased with the increases of artificial ageing times. The conductance and the soluble sugar of the seed extract increased with the increase of artificial ageing time,and had a significant negative correlation with germination percentage,germination energy,plumule length,root length and vigor index (P<0.01). However, the physiological change of seeds with endophyte fungi was relieved. This study suggested that the endophyte enhanced the seed germination of H. brevisubulatum with more plumule and root lengths,reduced the damage of plasmalemma and decreased the soluble sugar content and conductance of seed extract. Consequently, the deterioration of seed was reduced and the storage time of seed was extended.
