饲草型小黑麦遗传多样性的ISSR分析

ISSR Analysis on Genetic Diversity of Forage Triticale Genotypes

摘要:

本研究利用ISSR分子标记对30份饲草型小黑麦种质进行遗传多样性分析,以了解不同饲草型小黑麦种质间的遗传差异,为小黑麦种质资源保护、品种选育及分子鉴定提供理论依据。结果表明:参试小黑麦种质遗传多样性较丰富,利用 ISSR的12条引物共检测到124条扩增条带(位点),平均多态性比率(PPB)为77.42%。30份小黑麦种质的遗传相似系数(GS)为0.66~0.91。在GS=0.765处将30份小黑麦种质分为6类,其中8809及81S37与其他种质的遗传距离最大,单独聚为一类;826126和北联3号为第Ⅴ类;33RD ITSN、CHEROKEE和新小黑麦5号为第Ⅰ类;北联4号、PH389、DH796、中拉一号和安83-25为第Ⅳ类;其余种质聚为第Ⅱ类。第Ⅳ类与第Ⅴ类小黑麦种质的遗传相似系数均较小,如果其他性状符合育种目标,可相互进行杂交。

Abstract:

Genetic diversity of 30 forage triticale genotypes was investigated using ISSR-PCR molecular markers. This study would provide the theory basis for resources protection, breeding and molecular identification of forage triticale. The results indicated that the genetic diversity of 30 triticale genotypes was abundant. 124 bands were detected with 12 primers, and the proportion of polymorphic bands (PPB) was 77.42%. The genetic similarity coefficient for 30 triticale genotypes ranged from 0.66 to 0.91. The cluster analysis showed that the genetic similarity coefficient for boundaries was 0.765 and the 30 triticale genotypes were divided into 6 types. The genotype 8809 and 81S37 had the longest genetic distance and was clustered into different groups; genotypes 826126 and Beilian No.3 were clustered into the type Ⅴ; 33RD ITSN,CHEROKEE and New No.5 were clustered into the typeⅠ; the type Ⅳ included the genotypes Beilian No.4, PH389, DH796, Zhongla No.1 and An 83-25; the rest genotypes were clustered into the type Ⅱ. The genetic similarity coefficient of the 4th and 5th group was small so that they could be hybridized and breed new triticale varieties.
