三种葱莲属植物的倍性及FISH核型分析

Analysis of Ploidy Levels and FISH Karyotypes of Three Zephyranthes Cultivars

摘要: 本研究以5S rDNA为探针,利用FISH技术对3种葱莲属栽培品种有丝分裂中期染色体上的rDNA位点进行物理定位,分析3种葱莲属栽培品种的倍性及FISH核型。结果表明:3个品种存在两种倍性,其中‘Lily Pies’和‘Fragrance’为二倍体、‘King's ramson’为四倍体。它们的核型特点表现为:(1)‘Lily Pies’核型公式2n=2x=24=6 m+18 sm,染色体相对长度范围为6.17%~12.80%,属于2B型;(2)‘Fragrance’核型公式2n=2x=24=14 m+10 sm,染色体相对长度范围为6.36%~12.08%,属于2A型;(3)‘King's ramson’核型公式2n=4x=48=28 m+20 sm,染色体相对长度范围为5.68%~13.55%,属于2B型。3种葱莲属植物的FISH位点均位于染色体的长臂末端,分别为‘Lily Pies’含有4个5S rDNA位点,在6号和8号的两条染色上;‘Fragrance’含有3个5S rDNA位点,在两条6号染色体以及一条8号染色上;‘King's ramson’含有4个5S rDNA位点,分别在一条2号染色体、两条6号染色体和一条8号染色体上。本研究对葱莲属3个栽培品种的倍性和FISH核型的分析,将为该物种的遗传多样性分析和基因组结构解析提供新的信息。

Abstract: In this study,Fluorescence in situ hybridization (FISH) was used to physically locate the 5S rDNA repeat on the metaphase chromosomes of three Zephyranthes cultivars. The ploidy and FISH karyotype of 3 Zephyranthes cultivars were analyzed. The results showed that there were two types of ploidy in the three cultivars,among which ‘Lily Pies’ and ‘Fragrance’ were diploid and ‘King's ramson’ was tetraploid. The karyotype characteristics were as follows: ‘Lily Pies’ karyotype formula 2n=2x=24=6 m+18 sm,the relative length of chromosome ranged from 6.17% to 12.80%,and the karyotype belonged to 2B type. The formula of ‘Fragrance’ karyotype 2n=2x=24=14 m+10 sm,the relative length of chromosome was 6.36% 12.08%,and the karyotype belonged to 2A type. The formula of ‘King's ramson’ karyotype was 2n=4x=48=28 m+20 sm,the relative length of chromosome ranged from 5.68% to 13.55%,and the karyotype belonged to 2B type. The FISH loci of the three species were located at the end of the long arm of the chromosome,showing that ‘Lily Pies’ contained four 5S rDNA loci,which were on two homologous stains of No. 6 and No. 8,respectively. ‘Fragrance’contained three 5S rDNA loci,which were on two homologous chromosomes of No. 6 and one stain of No. 8. ‘King's ramson’ contained four 5S rDNA loci,which were on one on No. 2,two on No. 6 and one on No. 8. In this study,the ploidy levels and the FISH karyotypes analyzed in three cultivars of Zephyranthes would provide new information for the analysis of genetic diversity and genome structure of this species.
