沙打旺叶片愈伤组织诱导和植株再生的研究

Abstract: This paper is about study on the plantlet regeneration from leaf explants of Astragalus adsur gens.Five complete plantlets were finally obtained by the further transfering shoos onto the rooting medium.The MS medium was chosen adding to different concentrationes of 2, 4-D, NAA, IAA, BA, KT and ZT.A 85 % high frequent callus formed from one MS medium supplemented with 2,4-D(2mg/l), but the calli could not grow further, A lower frequent callus formed from MS medium with 2, 4-D(g/l) +ZT(0.1mg/l).These callus could grow further which could be subcultured and induced to form shoos.When the callus were put on to the medium (MS+0.7mg/l BA+0.2mg/l NAA), a lot of shoots appeared and callus did not grow.BA,NAA and 2,4-D had controlled the effect on the leaf of Astragalus adsurgens, and also effects of cane sugar and different media on callus formation.

摘要: 本文对沙打旺叶片外植体愈伤组织诱导和植株再生进行初步研究,建立了叶片外植体细胞无性系,并获得再生植株。试验以MS为基本培养基,附加不同浓度的2,4-D、NAA、IAA、BA、KT和ZT。单独附加2,4-D时,85%的外植体形成愈伤组织。2,4-D与ZT或BA配合使用时,形成愈伤组织的百分率虽降低,但这种愈伤组织可继代培养和诱导分化成苗。在MS培养基上附加BA和NAA时,则获得再生苗。BA、NAA和2,4-D对沙打旺叶片愈伤组织分化成苗有调节作用。此外,还探讨了蔗糖和不同培养基对愈伤组织形成的影响。
