RAPD Analysis on the Genomic DNA of Several Triticeae Grasses and Their Hybrids

几种小麦族禾草及其杂交后代基因组DNA的RAPD研究

Abstract: The Genomic DNA differences among Agropyron mongolicum ? Agropyrom cristatum cv.Fairway F1 hybrid,their backcross 1 (F1 hybrid ? A.mongolicum)and Elymus canadensis ? Hordeum brevisubulatum F1 hybrid were analyzed by using RAPD.The appropriate primers which can be used to determine the genetic differences among parents and their hybrids were selected.It was tested and verified thatthe genetic differences in DNA between E.canadensis and H.brevisubulatum were greater than that between A.mongolicum and A.cristatum cv.Fairway.The RAPD profiles of F1 hybrids tended to their motherobviously.RAPD can be used to identify hybrids and to study genetic breeding of grasses.

摘要: 应用随机扩增多态性DNA(RAPD)技术分析蒙古冰草、航道冰草及其种间杂种F1、BC1代和加拿大披碱草、野大麦及其属间杂种F1代的基因组DNA差异性。筛选出能检测各条本及杂交后代间遗传差异的适宜引物;从DNA分子水平验证出亲本加拿大披碱草和野大麦两属间种比亲本蒙古冰草、航道冰草二种间的亲缘关系远;杂种F1代的RAPD图谱有明显偏母本的倾向;随机扩增多态性DNA技术可用于牧草远缘杂种鉴定及遗传育种研究。
