Effects of Pb,Cd and Acid Stress on Seed Germination,Seedling Growth and Antioxidant Enzyme Activities of Paspalum wettsteinii

Pb、Cd和酸胁迫对宽叶雀稗种子萌发、幼苗生长及抗氧化酶活性的影响

Abstract: Soil acidification and heavy metal pollution are two major limiting factors for the ecological restoration of abandoned ion-type rare earth mining lands. Some special morphological and physiological mechanisms may exist in Paspalum wettsteinii due to its strong adaptation to the abandoned ionic rare earth mining lands in Changting,Fujian. Therefore,stress tests of different concentrations of Pb and Cd and different pH levels were designed to investigate their effects on seed germination,seedling growth,and antioxidant enzyme activities of Paspalum wettsteinii. The results showed that either high concentrations of Pb(≥ 500 mg·L-1) or Cd(≥ 50 mg·L-1) stress had significant inhibitory effects on Paspalum wettsteinii seed germination and seedling growth (P<0.05). Decreased pH values,pH-4.5,pH-5.5,had promoting effect on seed germination and seedling growth of Paspalum wettsteinii. The activity of catalase (CAT) and superoxide dismutase (SOD) in the early Pb treatment period and peroxidase (POD) activity in the middle and late Pb treatment periods increased first and then decreased. The activities of SOD and CAT in the early and late periods of Cd stress decreased first and then increased. With the decrease of pH values,the activity of SOD in the middle and late periods and the activity of POD in the early and middle periods both showed a trend of decrease first and then increase,the CAT activity in the early and late periods were the opposite. In summary,antioxidant systems showed the different responses to three environmental stresses in Paspalum wettsteinii. There were obvious differences in the germination time and SOD,POD and CAT activities.

摘要: 土壤酸化和重金属污染是限制离子型稀土矿废弃地植被恢复的主要因素,宽叶雀稗(Paspalum wettsteinii Hack)可在长汀离子型稀土矿废弃地正常生长,可能存在对酸和重金属胁迫特殊的适应机理。鉴于此,以宽叶雀稗为材料,设置不同浓度的Pb、Cd及酸胁迫试验,测定不同胁迫下宽叶雀稗种子萌发特性、幼苗生长及抗氧化酶活性的变化。结果表明,Pb (≥500 mg·L-1)、Cd (≥50 mg·L-1)胁迫对宽叶雀稗种子萌发、幼苗生长具有显著的抑制作用(P<0.05);pH-4.5,pH-5.5对宽叶雀稗种子萌发及幼苗生长有一定促进作用;Pb处理早期超氧化物岐化酶(SOD)、过氧化氢酶(CAT)活性及中后期过氧化物酶(POD)活性均呈先增后减趋势;Cd胁迫早期及晚期的SOD、CAT活性先减后增;酸处理中后期的SOD活性及早中期的POD活性均先减后增,早期及晚期的CAT活性则先增后减。宽叶雀稗抗氧化酶系统在Pb、Cd及酸胁迫下响应机制不同,SOD、POD、CAT活性高低有明显的时间差异和浓度效应。
