Agrobacterium rhizogenes-Mediated Transformation of Hairy Roots in Medicago ruthenica

发根农杆菌介导的花苜蓿毛状根转化体系的建立

Abstract: In order to develop an efficient method of transformation for Medicago ruthenica,Agrobacterium rhizogenes-mediated transformation of hairy roots was performed using M. ruthenica ‘Zhongke No.1’ as material. The incidence rate of hairy roots and the efficiency of transformation were analyzed. The results showed that,using this protocol,the transgenic hairy roots could emerge in about one week,and the incidence rate of hairy roots was 72%. The proportion of hairy roots in which genes were successfully transferred and over-expressed was 66.7%. This method of hairy root transformation provides a powerful tool to explore the functional genes in M. ruthenica and lay a foundation for alfalfa breeding.

摘要: 为开发一种高效的花苜蓿(Medicago ruthenica)转化体系,本试验以‘中科1号’花苜蓿为材料,使用发根农杆菌作为介导进行毛状根转化,并分析毛状根发生率和转化率。试验表明:利用本研究开发的方法可使花苜蓿在一周左右产生毛状根,其发生率可达72%;经鉴定,毛状根中成功转入基因并超表达的占66.7%。花苜蓿毛状根转化体系的建立将为其功能基因研究提供有力的手段,进而为苜蓿育种奠定基础。
