Abstract: TCP is a unique transcription factor in higher plants,paticipating in the regulation of biological processes such as,plant growth and development,aging,stress response,et al.. In this study we cloned ZjTCP20 gene from Zoysia japonica,investigated its characters through bioinformatics analysis,self-activation detection and expression pattern analysis. The complete coding region of ZjTCP20 gene was 624 bp,encoded 208 amino acids. ZjTCP20 protein was hydrophobic without signal peptide,and the phylogenetic tree analysis showed it closely similarity to ZmTCP20 protein of Zea mays. Subcellular localization revealed that the protein was resided in the chloroplast,cytoplasm and nucleus. Expression pattern of ZjTCP20 gene showed that it was highly expressed in young leaves and decreased expression with development,but three hormones including abscisic acid (ABA),methyl jasmonate (MeJA) and salicylic acid (SA) didn't have significant effects on its expression. Yeast self-activation experiment revealed that ZjTCP20 had an autoactivation activity. The study provides a scientific basis for further studies of the ZjTCP20 gene in Zoysia japonica.

摘要: TCP是高等植物所特有的一类转录因子,参与调控植物的生长发育、衰老及逆境应答等多个生物学过程。本研究从日本结缕草(Zoysia japonica)中克隆出ZjTCP20基因,并对该基因进行生物信息学分析、自激活检测、表达模式分析等来探究该基因特性。ZjTCP20基因完整编码区为624 bp,编码208个氨基酸。ZjTCP20蛋白无信号肽,具有疏水性,系统发育进化树结果显示其与玉米(Zea mays) ZmTCP20亲缘关系最近。亚细胞定位显示该蛋白定位于叶绿体、细胞质和细胞核上。ZjTCP20基因表达模式结果显示,该基因在幼嫩叶片中大量表达,随植物发育表达量逐渐减少,但茉莉酸甲酯(MeJA)等3个激素对基因的表达量没有显著影响。酵母自激活试验显示ZjTCP20具备自激活活性。本研究为进一步研究ZjTCP20基因提供了科学依据。
