Screening and Growth-promoting characteristics of Plant Growth-Promoting Rhizobacteria of Oat Rhizosphere in Alpine Reseeding Grassland of Zoige

若尔盖高寒补播草地燕麦根际促生菌的筛选及促生特性研究

Abstract: The Plant growth-promoting rhizobacteria (PGPR) were screened from the rhizosphere of oat in Zoige alpine reseeding grassland,which provided excellent the PGPR resources for production of microbial fertilizer suitable for the alpine reseeding grassland in Zoige. In this study,PGPR with different functions was screened by selective mediums,and the activity of nitrogen-fixing enzyme,phosphorus solubilization content and plant hormone secretion of the strain were measured. The classification status of the strain were determined by16S rDNA gene sequence and phylogenetic analysis,and growth promoting effect of the strain was tested by growth promoting test. The results show that 12 strains PGPR with excellent growth-promoting characteristics were initially screened from the rhizosphere of oat. Among them,7 strains had multiple growth-promoting characteristics,accounting for 58.33% of the total bacteria count,and were dominated by Pseudomonas;and 4 strains PGPR with the best growth-promoting effect were screened out. Among them,strains NCRS21 and PCRS14 had significant promoting effects on the oat growth (P<0.05);and strains NCHP9 and MCRS16 had significant promoting effects on the soil available phosphorus content (P<0.05). Therefore,Strains NCHP9,MCRS16,PCRS14 and NCRS21 have a stable growth-promoting ability,which have a potential application in the production of microbial fertilizer suitable for reseeding grassland in the Zoige alpine region.

摘要: 筛选若尔盖高寒补播草地燕麦根际的植物根际促生菌(Plant growth-promoting rhizobacteria,PGPR),可为生产适宜其微生物菌肥提供优良菌种资源。本研究采用选择性培养基筛选具不同功能的PGPR,并测定菌株的固氮酶活性、溶磷量、植物激素分泌量,16S rDNA基因序列与系统发育分析确定菌株的分类地位,促生试验检验菌株的促生效果。结果表明,从燕麦根际初步筛选出12株促生特性优良的PGPR,其中,7株具有多种促生特性,占总细菌数的58.33%,以假单胞菌属(Pseudomonas)为主,最终筛选出4株促生效果最好的PGPR,其中,菌株NCRS21、PCRS14对燕麦生长的促进作用显著(P<0.05),菌株NCHP9、MCRS16对土壤速效磷含量的促进作用显著(P<0.05)。因此,菌株NCHP9,MCRS16,PCRS14,NCRS21的促生能力稳定,在生产适宜若尔盖高寒补播草地的微生物菌肥方面具有应用潜力。
