四倍体‘紫光’狼尾草的诱导及鉴定分析

Abstract: In order to obtain tetraploid plants of Pennisetum alopecuroides ‘Ziguang’ and explore the optimal induction conditions, the young-ear of P.alopecuroides ‘Ziguang’ was used as material, and the induced embryogenic callus was used as explants in this study. The colchicine dipping method was used to induce mutagenesis of P. alopecuroides ‘Ziguang’.The induced tetraploid plants were identified through flow cytometry method (FCM) and root tip chromosome counting, and leaf stomatal characteristics and the phenotypic characteristics were compared between diploid and tetraploid plants. The results showed that the induction rate (55.56%) was the highest when the colchicine concentration was 0.10% and the treatment time was 48 hours.The results of FCM showed that the cellular DNA content of the induced tetraploid strain was doubled compared with that of the diploid plants. The root tip chromosome counting method found that the induced tetraploid plants(2n=4x=36) was twice the number of chromosomes in diploid plants (2n=2x=18).Through stomatal identification and comparison of plant phenotypic characteristics, tetraploid plants showed increased leaf width, decreased plant height, increased stomata, and decreased stomatal density compared with diploid plants.

摘要: 为获得‘紫光’狼尾草(Pennisetum alopecuroides ‘Ziguang’)四倍体植株并筛选最适诱导条件,本研究以‘紫光’狼尾草幼穗为试验材料,以其诱导出的胚性愈伤组织为外植体,采用秋水仙素浸渍法对胚性愈伤组织进行加倍处理,组织培养后获得再生植株。分别通过流式细胞术鉴定、根尖染色体计数法鉴定、气孔特征分析鉴定出诱导成功的四倍体植株,并对二倍体和四倍体植株的表型特征进行比对。结果表明:在秋水仙素浓度为0.10%,处理时间为48 h的条件下诱导率最高,可达55.56%;流式细胞术显示,四倍体植株的DNA相对含量为二倍体植株的2倍;根尖染色体计数法鉴定到四倍体植株(2n=4x=36)的根尖染色体数目是二倍体植株(2n=2x=18)染色体数目的2倍;通过气孔鉴定和植株表型特征比较,四倍体植株相较二倍体植株表现为叶宽增大、株高降低、气孔增大、气孔密度降低等特点。
