基于基因组Survey的翅果菊基因组大小评估及特征分析

Abstract: Lactuca indica is a plant species and valued for its medicinal, edible and forage properties. It demonstrates high biomass production and significant environmental adaptability, with considerable potential as a high-quality forage crop. However, the limited availability of genomic information on L. indica has hindered the effective use of its genetic resources and molecular breeding. In this study, we estimated the genome size and evaluated the genomic characteristics of L. indica using flow cytometry and genomic survey methods. Chromosome counting analysis confirmed the somatic chromosome number of 2n=18. Flow cytometry estimated the genome size of L. indica was between 4.26 and 4.50 Gb. Next-generation DNA sequencing technology and K-mer-based analysis further suggested that its genome size was approximately 4.76 Gb, with a heterozygosity of 0.24%. Repetitive sequences accounted for 73.76% of its genome. The preliminary assembly of the L. indica genome yielded a total length of 4.02 Gb with a contig N50 of 5070 bp. This study provided a preliminary genomic characterization of L. indica, laying the foundation for further molecular breeding and enhancement of forage traits.

摘要: 翅果菊(Lactuca indica)是一种兼具药用、食用及饲用价值的植物资源,其生物量大、适应性强,可作为优质饲草。目前,翅果菊的基因组信息仍比较匮乏,这限制了它的分子育种研究及资源利用。本研究利用流式细胞术和基因组Survey等实验手段,对翅果菊的基因组大小及特征进行综合评估。染色体计数分析结果显示,翅果菊体细胞染色体数目为2n=18;流式细胞术分析估测翅果菊基因组大小介于4.26~4.50 Gb区间;基于二代DNA测序的K-mer分析结果表明,翅果菊基因组大小约4.76 Gb,重复序列比例约73.76%,杂合率约0.24%;基因组初步组装后,获得了总长度为4.02 Gb,重叠群N50达5070 bp的初代翅果菊基因组。本研究提供了对翅果菊基因组的初步解析,为其未来分子育种及饲用价值挖掘提供了数据支持。
