杂草致病菌株粉红螺旋聚孢霉JZ-28的分离、鉴定及除草潜力评价

Abstract: To screen biocontrol strains with high herbicidal potential, the pathogenic fungus JZ-28 was isolated from naturally infected leaves of Viola philippica in Qinghai Province. Through morphological characterization and multi-gene (ITS, EF-1α, LSU) phylogenetic analysis, the isolate was identified as Clonostachys rosea. Pathogenicity assays demonstrated potent herbicidal activity of strain JZ-28, inducing chlorosis in detached leaves of four dominant broadleaf weed species from the Qinghai-Xizang Plateau(Malva verticillataElsholtzia densa Senecio vulgaris, and Chenopodium album)within 7 days post-inoculation (dpi). Disease incidence rates in whole-plant assays reached 83%, 95%, 100%, and 70%, respectively. Safety evaluation on six major crops and three cool-season vegetables in Qinghai revealed that JZ-28 was safe for four crops: Zea maysVicia fabaPisum sativumBrassica napus, and three vegetables: Cucumis sativusSolanum lycopersicum, and Solanum melongena. Lightly Susceptible was observed in highland barley Hordeum vulgare and Triticum aestivum. These findings indicated JZ-28’s potential as a broad-spectrum bioherbicide against broadleaf weeds in multiple cropping systems. Culture optimization identified sucrose and soybean meal as optimal carbon and nitrogen sources for enhancing mycelial growth and sporulation. This study provided the first evidence of Clonostachys rosea’s bioherbicide potential in alpine ecosystems, addressing a critical research gap in microbial herbicide development and offering sustainable weed management solutions for fragile environments.

摘要: 为筛选具有高效除草潜力的生防菌株,本研究从青海省自然感病紫花地丁(Viola philippica)叶片中分离出致病菌JZ-28。通过形态学及多基因 (ITS/EF-1α/LSU) 联合鉴定,确定为粉红螺旋聚孢霉(Clonostachys rosea)。致病性试验表明其具有强效除草活性,JZ-28在7 d可使青藏高原4种优势阔叶类杂草冬葵(Malva verticillata)、密花香薷(Elsholtzia densa)、欧洲千里光(Senecio vulgaris)和藜(Chenopodium album)的离体叶片黄化发病,活体植株试验发病率分别为83%,95%,100%,70%。评价JZ-28对青海省6种主栽作物和3种冷凉蔬菜的安全性,表明该菌株对玉米(Zea mays)、蚕豆(Vicia faba)、豌豆(Pisum sativum)、油菜(Brassica napus)4种作物和黄瓜(Cucumis sativus)、番茄(Solanum lycopersicum)、茄子(Solanum melongena)3种冷凉蔬菜表现为安全,对青稞(Hordeum vulgare)、小麦(Triticum aestivum)表现为轻微感病。培养条件优化试验表明,蔗糖与大豆粉分别为最佳碳、氮源,可促进菌丝生长与孢子产量提高。本研究首次揭示了粉红螺旋聚孢霉作为高寒地区生物除草剂的潜力,为脆弱生态系统的杂草可持续管理提供了新的途径。
