芍药种子层积过程中生理生化变化研究

Abstract: The accumulation process of Paeonia lactiflora seed lays the foundation for studying its dormancy mechanism by exploring its morphological changes,nutrient substances,enzyme activity,endogenous hormones,and the expression of key genes. The study investigated the morphological changes of seeds and measured the levels of nutrient substances,enzyme activity,hormones,and key genes expression. The process of dormancy release was divided into lower embryo axis growth,embryo bud swelling,and upper embryo axis growth;initial storage materials were primarily composed of fats,accounting for about 28%. During the dormancy release process,fats,starch,proteins,and soluble sugars decreased overall,providing substances for germination. Fats were consumed the most during dormancy release,serving as the main source of energy substances. The changes in enzyme activity during the process of dormancy release reflected the trend of nutrient changes,with an overall enhancement in antioxidant enzyme activity. During dormancy release,there was an initial increase,followed by a decrease in gibberellic acid (GA) content,while the content of abscisic acid (ABA) and the GA/ABA ratio significantly decreased. Changes in key genes aligned with changes in endogenous hormone levels. The release of dormancy in Paeonia lactiflora seeds required fluctuations of temperature from warm to cold conditions,enhanced enzyme activity,and the breakdown of nutrients to provide energy for dormancy release. The significant decrease in ABA content was the main hormone inhibiting germination. Releasing dormancy in Paeonia lactiflora seeds required coordinated regulation of energy metabolism and hormones.

摘要: 芍药(Paeonia lactiflora)种子层积过程中形态、营养物质、酶活性、内源激素及关键基因的表达,为其休眠机制研究奠定基础。研究种子的形态变化,测定营养物质、酶活性、激素及关键基因的表达。休眠解除过程分为下胚轴生长、胚芽膨大和上胚轴生长;初始储藏物质以脂肪为主,约28%。休眠解除过程脂肪、淀粉、蛋白质和可溶性糖整体下降,为萌发提供物质。脂肪在休眠解除中消耗最多,为主要供能物质。休眠解除过程分解营养物质的酶活性变化符合营养物质变化趋势,抗氧化酶活性整体增强;休眠解除过程中GA含量呈现上升-下降,ABA含量和GA/ABA比值显著下降,关键基因变化符合内源激素含量变化。芍药种子休眠解除须满足暖温-低温的变化,酶活性增强,营养物质分解为休眠解除提供能量,ABA含量显著下降是抑制萌发的主要激素。芍药种子休眠的解除需要能量代谢和激素共同调控。
