EMS处理对老芒麦种子萌发和生理生化指标的影响

Abstract: To determine the optimal EMS mutagenesis system for ‘Tongde’ Elymus sibiricus， this study utilized ethyl methanesulfonate （EMS） seed soaking treatments including five concentration levels （0%， 0.4%， 0.8%， 1.2% and 1.6%） and three exposure durations （12， 16 and 20 h）. Germination rate， germination potential， survival rate， and key physiological and biochemical indicators were measured to comprehensively evaluate the effects of EMS treatments on seed vigor and physiological responses. The results demonstrated that germination rate， germination potential， and greenhouse survival rate decreased significantly with increasing EMS concentration and treatment duration. Additionally， the interaction between EMS concentration and exposure time significantly influenced endogenous hormone levels， enzyme activities， and malondialdehyde （MDA） content. Based on the germination rate， germination potential， and greenhouse survival rate， the optimal EMS mutagenesis system for ‘Tongde’ Elymus sibiricus was determined to be 0.4% EMS treatment for 16 hours. This study provided a theoretical foundation and technical support for chemical mutagenesis breeding and the development of new Elymus sibiricus varieties.

摘要： 为确定‘同德’老芒麦（Elymus sibiricus）的最佳诱变体系，本研究采用甲基磺酸乙酯（Ethyl Methanesulfonate，EMS）对种子进行浸泡处理，设置了5个浓度梯度（0%，0.4%，0.8%，1.2%，1.6%）和3个处理时间梯度（12，16和20 h），通过测定种子发芽率、发芽势、存活率及主要生理生化指标，系统解析了不同EMS处理对种子活力和生理生化的影响。结果表明，随着EMS浓度和处理时间的增加，种子发芽率、发芽势及温室存活率显著下降；不同浓度与时间的交互作用对内源激素含量、酶活性及丙二醛含量产生显著影响。综合发芽率、发芽势和温室存活率，确定了‘同德’老芒麦的最佳诱变体系为0.4% EMS处理16 h。研究为老芒麦的化学诱变育种及新品种选育提供了理论依据和技术支撑。
