南疆沙区油莎豆根际耐盐促生菌株的筛选

Abstract: To screen salt-tolerant and growth-promoting bacterial strains adapted to the extreme environment of Southern Xinjiang from the rhizosphere soil of Cyperus esculentus L. and enrich the strain resource bank for saline-alkali sandy soils in Xinjiang, this study employed the targeted screening medium culture method, shake flask culture method, and 16S rDNA identification method to isolate salt-tolerant and growth-promoting strains, determine their growth-promoting characteristics, and finally further verify the growth-promoting effects of these strains through Cyperus esculentus seed germination tests and potted seedling experiments. A total of 10 salt-tolerant and growth-promoting strains were obtained, which belonged to the genera BacillusVreelandellaExiguobacterium, and Bisbaumannia, with their salt tolerance thresholds ranging from 4% to 8% NaCl. Among these strains, strain Y-38 (Exiguobacterium acetylicum) exhibited prominent inorganic phosphorus-solubilizing ability (0.65 mg·L-1), strain Y-35 (Exiguobacterium mexicanum) showed the highest production of indole-3-acetic acid (IAA) (64.35 mg·L-1), and strain Y-46 (Bisbaumannia pacifica) could tolerate 8% NaCl. In the seed germination experiment, the germination rate of the Y-38 treatment group reached 82.00%, which was significantly 26.89% higher than the control group (P<0.05); in the potted experiment, growth indicators of the Y-38 treatment group, including plant height (39.17 cm), leaf width (5.22 mm), and stem diameter (5.52 mm), were all significantly higher than those of the Y-46 and Y-35 treatment groups (P<0.05). The comprehensive ranking of the growth-promoting effects of the three strains was Y-38>Y-46>Y-35. Ultimately, the high salt tolerance and growth-promoting function of strain Y-38 (Exiguobacterium acetylicum) was confirmed, providing a bacterial strain resources for the improvement of saline-alkali soils.

摘要: 为从油莎豆(Cyperus esculentus L.)根际土壤中筛选适应南疆极端环境的耐盐促生菌株,丰富新疆盐碱沙土地菌株资源库,本研究采用定向筛选培养基培养法、摇瓶培养法和16S rDNA鉴定法,对耐盐促生菌进行筛选,并测定其促生特性。最终通过油莎豆种子萌发和盆栽幼苗试验,进一步验证其促生效果。本研究共获得10株耐盐促生菌,分属于芽孢杆菌属(Bacillus)、弗里兰氏菌属(Vreelandella)、微小杆菌属(Exiguobacterium)和比斯鲍曼氏菌属(Bisbaumannia),耐NaCl阈值范围为4%~8%。其中菌株Y-38(Exiguobacterium acetylicum)溶无机磷能力突出(0.65 mg·L-1),Y-35(Exiguobacterium mexicanum)产吲哚-3-乙酸(Indole-3-acetic acid,IAA)量最高(64.35 mg·L-1),Y-46(Bisbaumannia pacifica)耐受8%NaCl。种子萌发试验中,Y-38处理组发芽率达82.00%,较对照显著提高26.89%(P<0.05);盆栽试验中Y-38处理组的株高(39.17cm)、叶宽(5.22mm)、茎粗(5.52mm)等指标均显著高于Y-46和Y-35处理组(P<0.05)。三株菌的促生效果通过综合排序为:Y-38>Y-46>Y-35,最终确定了Y-38(乙酰微小杆菌,Exiguobacterium acetylicum)的高耐盐性与促生功能,为盐碱土壤改良提供了菌种资源。
