根际促生菌的定殖及其对不同温度下紫花针茅生长的影响

Abstract: To screen plant growth-promoting rhizobacteria (PGPR) that are capable of stably colonizing the rhizosphere of Stipa purpurea, four PGPR strains previously isolated from this alpine grass were evaluated. Rifampicin-resistant derivatives were generated to monitor root-zone colonization dynamics, and the effects of bacterial inoculation on seed germination, seedling morphology, and physiological indices were assessed under the culture of 15℃, 20℃, and 25℃. Serratia proteamaculans GD-1-1 and Paenarthrobacter nitroguajacolicus YD-2-4 established stable populations in the S. purpurea rhizosphere, and the colonization amounts were 1.22×108 and 0.06×108 CFU·g-1 at 20 d, respectively. Dilutions of YD-2-4 (1×101 and 1×102 CFU·mL-1) significantly enhanced germination and seedling growth across 15℃-25℃, concurrently increasing antioxidant enzyme activities and lowering malondialdehyde content. In contrast, the stimulatory effect of GD-1-1 (1×102 and 1×103 CFU·mL-1) declined with temperature and disappeared at 15℃. These results indicated that YD-2-4 possessed robust colonization ability and broad temperature adaptability, making it an excellent candidate for the development of microbial inoculants for alpine grasslands.

摘要: 为筛选能有效定殖于紫花针茅根际的促生菌(PGPR),本研究以4株分离自紫花针茅根际的PGPR为候选菌株,通过利福平抗性筛选检测其根际定殖动态,并在15℃,20℃和25℃培养条件下,测定接种PGPR对紫花针茅发芽率、形态及生理指标的影响。结果显示,变形斑沙雷氏菌(Serratia proteamaculans)GD-1-1和硝基愈创木酚类节杆菌(Paenarthrobacter nitroguajacolicus)YD-2-4能有效定殖于紫花针茅根际,20 d时的定殖量分别为1.22×108和0.6×107 CFU·g-1。菌株YD-2-4的1×101和1×102 CFU·mL-1稀释液,在15℃~25℃范围内均能显著促进种子发芽和幼苗生长,同时提高植株抗氧化酶活性、降低丙二醛含量;而菌株GD-1-1的1×102和1×103 CFU·mL-1稀释液的促进作用随温度降低而减弱,15℃下无显著促进效果。说明菌株YD-2-4根际定殖能力稳定且温度适应范围广,可作为高寒草地微生物菌剂开发的优良菌株。
