鲁梅克斯k-1杂交酸模及野生酸模遗传多态性分析

Abstract: The genetic polymorphisms of three continued generations of Rumex k-1 (R. patientia×R.tianschanicus cv.Rumex k-1) and several wild Rumex spp. were analysed, using random amplified polymorphic DNA markers(RAPD). The result shows that there are no different polymorphic loci among the three successive generations of Rumex k-1, showing genetic stability, except for the 12 plants of the certified seed of Rumex k-1; while among the different populations of the same wild Rumex species growing in different sites, there appear abundant genetic polymorphisms loci. Seven primers selected from OPA twenty primers amplifies 45 RAPD fragments. The length of most of the amplified fragments range from 300 bp to 2500 bp.

摘要: 利用RAPD技术对鲁梅克斯k-1杂交酸模的3个繁育世代和野生酸模进行遗传多态性分析,结果表明:3个世代之间不存在多态性位点差异,3个世代是稳定遗传的;不同种和不同地点的同一个野生种之间存在着多态性位点差异;在合格种的12个单株间具有明显的遗传多态性;采用混合取样可消除品种内的个体遗传差异性,较好地代表群体遗传特征;从0PA组选出7个能扩增出清晰且稳定带谱的引物,7个引物共检测出4个位点,DNA片段的大小在300~200bp之间.
