Zymogram Analysis of Different Sexual Buchloe dactyloides

不同性别野牛草同工酶酶谱分析

Abstract: Superoxide dismutase (SOD), peroxidase (POD) and polyphenol oxidase (PPO) isozymes of leaves and spikes from different sexual Buchloe dactyloides (male, female or monoecious) specimens were analyzed using polyacrylamide gel electrophoresis. Results showed that the POD and PPO zymogram of spikes from male plants had distinct bands, Rf0.12 and Rf0.15, respectively; the PPO zymogram of both leaves from female plants and spikes from monoecious plant had a special band, Rf0.12. Therefore, foliar POD and PPO zymogram can be used for sex determination during early growth stages of Buchloe dactyloides.

摘要: 利用聚丙烯酰胺垂直板凝胶电泳技术对3种性别野牛草(Buchloe dactyloides)的幼叶和幼穗的过氧化物酶(POD)、超氧化物歧化酶(SOD)和多酚氧化酶(PPO)同工酶酶谱进行分析,以期通过酶谱的差异表达对野牛草进行早期性别鉴定。结果表明:雄株野牛草幼穗的POD和PPO同工酶各有1条特异条带Rf0.12和Rf0.15;雌雄同株的幼穗PPO同工酶酶谱中有1条特异条带Rf0.12;雌株的叶片PPO同工酶酶谱有Rf0.12的特异性条带。通过分析上述3种同工酶的酶谱发现,叶片的POD和PPO同工酶酶谱可以用来对野牛草进行早期性别鉴定。
