Cloning,Expression Characteristics and Subcellular Localization of PvJAZ1 Gene from Switchgrass

柳枝稷PvJAZ1基因的克隆、表达特性与亚细胞定位

Abstract: JAZ protein plays significant roles in plant growth and development and stress signaling pathways. In order to explore the regulatory mechanism of JAZ protein in switchgrass (Panicum virgatum L.),PvJAZ1 gene was cloned from switchgrass,and its molecular characteristics,expression characteristics and subcellular localization were analyzed. The results are as follows:The open reading frame of PvJAZ1 is 630 bp and encodes 209 amino acids. It is an unstable hydrophilic protein. The protein contains the conserved TIFY and Jas domains of the JAZ subfamily. Phylogenetic analysis indicated that PvJAZ1 was closely related to JAZ protein of Panicum miliaceum and Panicum hallii. Promoter analysis shows that the promoter region of PvJAZ1 gene contains cis-acting elements related to light response,hormone response,stress response and growth process;PvJAZ1 responds strongly to IAA,salt and drought stress,and its expression level in spikelet was significantly higher than that in other tissues;PvJAZ1 protein is located on the nucleus and endoplasmic reticulum. This research preliminarily determined that switchgrass PvJAZ1 gene plays critical roles in hormone,stress signal response and growth and development processes,laying a foundation for the exploration of PvJAZ1 gene function.

摘要: JAZ蛋白在植物生长发育和胁迫信号通路中具有关键作用。为探究JAZ蛋白在柳枝稷(Panicum virgatum L.)中的调控机制,本研究从柳枝稷中克隆了PvJAZ1基因,并对其分子特征、表达特性与亚细胞定位进行了分析。结果研究表明:PvJAZ1的开放阅读框长度为630 bp,编码209个氨基酸,为不稳定的亲水性蛋白,该蛋白含有JAZ亚家族保守的TIFY与Jas结构域;系统进化分析表明PvJAZ1蛋白与糜子(Panicum miliaceum)和哈氏黍(Panicum hallii)的JAZ蛋白亲缘关系较近;启动子分析表明PvJAZ1基因启动子区域包含与光响应、激素响应、胁迫响应和生长进程相关的顺式作用元件;PvJAZ1在生长素(auxin,IAA)、盐和干旱胁迫下反应强烈,在小穗中的表达量显著高于其它组织;PvJAZ1蛋白定位于细胞核和内质网上。本研究初步确定柳枝稷PvJAZ1基因在激素、胁迫信号应答和生长发育进程中具有关键作用,为探究PvJAZ1基因功能奠定了基础。
