青海草原毛虫IR基因的鉴定及组织表达分析

Abstract: In this study,based on the transcriptome data of Gynaephora qinghaiensis,bioinformatics methods and qRT-PCR technology were used to identify and analyze the expression profiling of GqinIRs genes. In this study,a total of 12 GqinIRs genes were identified,and bioinformatics analysis showed that the IRs of Gynaephora qinghaiensis belonged to amphoteric proteins,except for GqinIR7,GqinIR8a,GqinIR10a,GqinIR75a,and GqinIR76b,all of them had signal peptides and 2~4 transmembrane domains,and the second- and third-order structures were mainly composed of α helix and random coils. Phylogenetic results showed that the IR of Gynaephora qinghaiensis was closely related to Oriental armyworm and Spodoptera exigua. Gene expression profile analysis showed that the relative expression levels of the 12 IR genes in different tissues of Gynaephora qinghaiensis were different,and the GqinIR6 gene was basically not expressed in the antennae of the male,while the remaining 11 GqinIRs genes were expressed in the antennae of the male,while their expression levels were different (P<0.05). This study provides an important basis for further study of the recognition and response mechanism of ionotype receptor proteins of Gynaephora qinghaiensis to external odor molecules,and lays a foundation for further research on the olfactory mechanism of Gynaephora qinghaiensis.

摘要: 本研究利用生物信息学分析及qRT-PCR技术,对GqinIRs基因进行鉴定及表达谱分析。本研究共鉴定出12个GqinIRs基因,生物信息学分析发现青海草原毛虫IRs属于两性蛋白,除GqinIR7、GqinIR8a、GqinIR10a、GqinIR75a、GqinIR76b都具有信号肽,具有2~4个跨膜结构域,二三级结构主要由α螺旋和无规则卷曲组成;系统进化表明青海草原毛虫IR与东方黏虫和疆夜蛾亲缘关系较近。基因表达谱分析表明,鉴定的青海草原毛虫12个IR基因在雌雄不同组织体现不同的表达水平(P<0.05)。本研究为进一步研究青海草原毛虫离子型受体蛋白对外界气味分子的识别及反应机制提供重要依据,并且为深入研究青海草原毛虫嗅觉机制奠定基础。
