Screening and Evaluation of Salt-tolerant Germplasm of Sainfoin at Seedling Stage

红豆草苗期耐盐种质鉴定及综合评价

Abstract: To evaluate the salt tolerance of sainfoin (Onobrychis viciaefolia), a study was conducted using 27 sainfoin materials from domestic and international sources. The variations in related morphological and physiological indices in response to a 0.8% NaCl treatment were analyzed. The results showed that under salt stress, several parameters, including plant height, leaf area, root-shoot ratio, leaf relative water content and K+ concentrations, decreased in the tested sainfoin. Conversely, root activity, superoxide anion (O2-) content, hydrogen peroxide (H2O2) content, proline (Pro) content, malondialdehyde (MDA) content, catalase (CAT) activity, peroxidase (POD) activity, superoxide dismutase (SOD) activity, Na+ and Ca2+ contents increased. Comprehensive evaluations using membership function and cluster analysis revealed that sainfoin varieties ‘1994’ and ‘13523’ exhibited strong salt tolerance, while ‘2208’, ‘Mengnong’, and ‘6’ displayed the lowest tolerance levels. Regression analysis identified plant height, Ca2+ content, O2- content, leaf area, leaf relative water content, root activity, and POD activity as the primary indicators for assessing salt tolerance in sainfoin seedlings. The findings of this study contributed valuable germplasm for the breeding of new salt-tolerant sainfoin varieties.

摘要: 为筛选耐盐性优异的红豆草(Onobrychis viciaefolia)材料,以国内外27份红豆草为研究对象,探讨其相关形态和生理指标在0.8% NaCl处理下的响应差异。结果表明,盐胁迫下,供试红豆草除株高、叶面积、根冠比、叶片相对含水量与K+含量均呈降低趋势外,根系活力、超氧根阴离子(Superoxide anion,O2-)含量、过氧化氢(Hydrogen peroxide,H2O2)含量、脯氨酸(Proline,Pro)含量、丙二醛(Malondialdehyde,MDA)含量、过氧化氢酶(Catalase,CAT)活性、过氧化物酶(Peroxidase,POD)活性、超氧化物歧化酶(Superoxide dismutase,SOD)活性、Na+及Ca2+含量均呈升高趋势。隶属函数和聚类分析综合评价表明,材料‘1994’和‘13523’耐盐能力强,材料‘2208’‘蒙农’红豆草及‘6’红豆草耐盐能力最差。回归分析表明,株高、Ca2+含量、O2-含量、叶面积、叶片相对含水量、根系活力和POD活性可作为红豆草苗期耐盐性评价首选指标。研究结果为耐盐红豆草新品种选育提供了优异的种质材料。
