耐盐苜蓿与敏盐苜蓿RAPD多态性研究

Studies on RAPD Polymorphic in Salt-Tolerant Alfalfa and Salt Sensitive Alfalfa

摘要: 以耐盐苜蓿和敏盐苜蓿为材料,通过RAPD反应,对8组的160条引物进行筛选.结果表明,经过5轮筛选,OPA组6条引物、OPD组9条引物、OPE组6条引物、OPF组7条引物、OPH组5条引物、OPC组9条引物、OPG组2条引物、OPN组7条引物共51条都可作为多态性引物,其中以OPA-17、OPD-5、OPD-7等3条引物的DNA多态性最稳定.在"中苜一号”DNA组上,OPA-17可在2200bp处产生一条多态性谱带,并在380bp出现谱带缺失现象.OPD-5可在3500bp处形成一条清晰度极高的谱带,OPD-7则在650bp处无谱带产生.这3条引物所检测出的耐盐苜蓿与敏盐苜蓿具有稳定的DNA多态性,该多态性与其耐盐性状有关.

Abstract: The salt tolerant alfalfa and the salt sensitive alfalfa are used as material of extracting DNA,through RAPD reaction,we can select the primer of polymorphic DNA from 160 lines in eight groups' primers.After five turns,51 lines being of polymorphic DNA have been found.Among them OPA 17 OPD 5 and OPD 7 are three lines of the most stable polymorphic DNA.On the DNA of Medicago sativa L.“Zhongmu NO.1”,OPA 17 is able to produce a band of polymorphic DNA in 2200 bp DNA size and lose a band in the 380 bp;OPD 5 can lead a clear band in the 3500 bp; OPD 7 has lack of the band in 650 bp DNA point.The polymorphic DNA having stable is inspected by the three lines of primer.They maybe relact with the salt tolerance in Medicago sativa L.“Zhongmu NO.1”.
