柠条锦鸡儿DNA提取及AFLP反应体系的建立

摘要: 以柠条锦鸡儿(Caragana korshinskii)种子为材料,利用改进的酚一氯仿法,提取到高质量的DNA,建立柠条锦鸡儿AFLP银染反应体系。结果表明:首次在柠条锦鸡儿DNA提取、酶切连接、预扩增和选择性扩增等实验过程取得成功;得到清晰的柠条锦鸡儿AFLP指纹图谱;为建立柠条锦鸡儿基因库、品种选育以及亲缘关系的研究提供参考。

Abstract: The extraction of DNA with phenol-cholorofore method from Caragana korshinskii seed and AFLP fingerprint profile of C. korshinskii were studied. The high resolution AFLP fingerprints were clear,showing the success in the DNA extraction, restriction-ligase reaction,pre-amplification and silver staining techniques.The results could supply references to the gene pool establishment,selection and breeding of cultivars and genetic relationships in C. korshinskii.
